Objective: This study observed the effects of Grape seed proanthocynadin extract (GSPE )on the expressions of PPARγmRNA ?and the activities of the downstream molecule- NF-кB of thoracic aorta of the model SD rats fed with a high cholesterol and GSPE diet. Pioglitazone is positive to procyandins. Furthermore,the effects of Procyanidins on the levels o?f blood fat of the model rats were investigated.Methods: Four groups were randomly selected from 32 male Sprague-Dawley rats: control group(C), the hyperlipemia group(HL highlipid diets), the pioglitazone group(PI highlipid diets) , the procyanidin group(PC highlipid diets). The PI is positive to PC. All groups were fed a high cholesterol diet for six weeks, At the same time, rats were treated with either 10 mg/kg.d body weight pioglitazone ,100 mg/kg.d body weight procyanidins or distilled water by gavage daily till the end of the experiment. At 0 week, 3th week,6th week, blood was gotten by tail vein. Rats were sacrificed at 6th week, and thoracic aorta were gotten and reserved by -70℃. The levels of serum TG, TC, HDL-C were tested by kits, the expressions of PPARγmRNA were measured by RT-PCR and the activities of NF-кB by EMSA.Results:1. The levels of blood fats in HL:At the 3th week, 6th week,the levels of TG,TC in HL rats is higher than C rats (p<0.05).2. The expressions of PPARγmRNA and the activities of NF-кB of thoracic aorta of SD rats:(1)Compared with C rats, the levels of PPARγmRNA of thoracic aorta were lower, the activities of the NF-кB of thoracic aorta in HL were higher (p<0.05), and showed to be statistical difference (p<0.05).(2) Compared with HL rats, the levels of PPARγ?mRNA of thoracic aorta were improved and the activities of the NF-кB of thoracic aorta were reduced in PC rats(p<0.05). Compared with PI,C rats, the levels of PPARγ?mRNA of thoracic aorta and the activities of the NF-кB of thoracic aorta in PC rats had no statistical difference.3.At the 6th week,Compared with HL rats, the levels of TG,TC in PC rats (p<0.05) were reduced, but had no statistical difference between PI,C and PC rats.Conclusion:1.Procyanidins can prevent the levels of TG,TC improving in accordance with reports.2.Procyanidins can prevent the expressions of PPARγmRNA of thoracic aorta reducing.3.Procyanidins can prevent the of NF-кB of thoracic aorta activating.
