Objective: 1.To investigate the association between single nucleotide polymorphisms(SNP),named as rs6832151 in the 4p14 and Graves’ disease(GD) in the Chinese Han population from Bengbu,Anhui.2.To Further investigate the association between single nucleotide polymorphisms(SNP),named as rs6832151 in the 4p14 and GD patients clinical features,including sex,age, TRAb levels, Graves’ ophthalmopathy, degree of goiter.Methods: 1.In 2007-2009 in Bengbu Medical College in the first affiliated hospital of Bengbu endocrine clinic patients, select the group of 617 GD patients and 4915 healthy controls, collected fresh blood, and DNA extraction blood cells,Case group 617 patients consistent with guide of diagnosis and treatment of thyroid disease in China about the diagnostic criteria of GD;2.With Taq Man probe technique, testing 617 GD patients and 4915 healthy controls of rs6832151 in the 4p14 and the genotype of GD patients and normal controls werecorrelation analysis, the relationship between SNPs and GD susceptibility;3.Analysis of SNP- rs6832151 clinical manifestations of the differences between different genotype subgroups, analysis the correlation between SNPs and clinical indicators.Result: 1.The genotypes of Rs6832151 in 4p14 was GG、GT、TT.The GG genotype frequency of Rs6832151 in GD group was higher than control group,respectively 17.99% vs 11.96%.The G genotype frequency of Rs6832151 in GD group was higher than control group,respectively 42.70% vs 34.87%.Rs6832151_G in 4p14 was strongly associated with GD(χ2=29.29, P= 6.22×10-8, OR=1.39);2.Three genetic models(additive model 、 dominant model 、 recessive model)built according to the locus genotyping(GGvs GTvs TTGG+GTvs TTGGvs GT+TT)were statistically significan(χ2=26.69,P=3.57×10-7;χ2=21.04,P=4.50×10-6;χ2=18.04,P=2.17×10-5);3.Clinical characteristics such as sex,age, TRAb levels, Graves’ ophthalmopathy, degree of goiter difference between the genotype subgroups were not statistically significant(P>0.05).Conclusion: 1.Rs6832151_G in 4p14 is Graves’ disease of Bengbu population susceptibility genes,and is related to the high risk of GD.2.Clinical characteristics such as sex,age, TRAb levels, Graves’ ophthalmopathy, degree of goiter difference between the genotype subgroups were not statistically significant...
