Background Tuberculous meningitis is one kind of the central nervous systeminfection,with high morbidity、 mortality,and poor prognosis,but there is still noeffective diagnostic tool for clinical work. The research about determination cytokines incerebrospinal fluid is relatively small in terms of tuberculous meningitis, and there is norelevant report about screening lots of cytokines in cerebrospinal fluid of tuberculousmeningitis both at home and abroad.Objective To select cytokines in cerebrospinal fluid from tuberculous meningitis forprobable diagnosis.Methods120kinds of cytokines were detected with protein chips, and the results werecompared among2tuberculous meningitis patients、2viral meningitis patients and2noninfectious neurologic disease patients to select the differential cytokines which weredetected among32persons with tuberculous meningitis、18persons with viral meningitisand18persons noninfectious neurologic disease by ELISA again to estimate the applicationvalue.Results1、83kinds of cytokines were found by protein chip, and6kinds of differentialcytokines were detected, which were respectively. Monocyte chemoattractant protein-2(MCP-2)、Macrophage inflammatory protein-1beta(MIP-1β)、Macrophage inflammatoryprotein-1alpha(MIP-1α)、Interferon gamma(IFN-γ)、Monocyte chemoattractant protein-3(MCP-3)and Interleukin-8(IL-8). 2、The results about6kinds of differential cytokines detected by ELISA showed that: theMIP-1β、 MCP-2、 MIP-1α、 IFN-γ、 MCP-3and IL-8levels in the samples from thetuberculous meningitis and viral meningitis patients were significantly higher than those fromnoninfectious neurologic diseases(P<0.05).The contents of MCP-2(6.5±6.3pg/ml and1.5±0.5pg/ml)、IFN-γ(253.3±182.5pg/ml and8.5±2.9pg/ml)、 IL-8(37134.8±27192.7pg/ml and16805.8±20108.6pg/ml) were significantly higher in patients with viralmeningitis(P<0.05), while there were no significant difference in MIP-1β、MIP-1α、MCP-3between the samples from the tuberculous meningitis and the viral meningitis(P>0.05).Conclusion1.The protein chip can preliminarily select the differential cytokines in thecerebrospinal fluid from tuberculous meningitis;2. The ELISA method can help to select the differential cytokines again;3.MCP-2、IFN-γ、IL-8in the cerebrospinal fluid are closely related with tuberculousmeningitis.
