Objective:To investigating whether syncytiotrophoblast microparticles(STBM)from different pregnant women can induce preeclampsia-like changes inpregnant rat. To provide a basis for the pathogenesis of preeclampsia by investigatingwhether STBM could cause the maternal vascular endothelial dysfunction andinflammation in vivo.Methods:10cases with severe preeclampsia and10cases of normalpregnancies as control were recruited.STBM were prepared from the placentas whichwere collected immediately after elective caesarean section,by modification of themethod of Smith et al.Different sources of STBM(STBM from preeclampsia women andnormal pregnancies) and normal saline were injected into rat tail veins. The systolicblood pressure(SBP),24hr urine protein,hepatic and renal function were comparedamong the different groups.The kidney biopsy was observed by light microscopy afterHE staining and PAS staining。The level of NO、ET-1、TNF-αand IL-6in serum wereevaluated by ELISA。The placentas and fetus were weighed after dissected.Results:①The SBP of the rats in the preeclampsia group(P group)(191.7±8.3mmHg)were significantly higher than the normal group(N group)(123.4±5.1mmHg)and the saline group(S group)(116.7±48mmHg)(P<0.01)。 The N group were higher than S group(P<0.05)。②The24hr urine protein of the ratsin P group(13.58±6.50mg/24h)were significantly higher than N group(4.02±1.42mg/24h)and S group(2.88±0.57mg/24h)(P<0.01).The N group and S groupwere simila(rP>0.05).③The plasma albumin of the rats in P group(31.90±1.60g/L)were significantly lower than the N group(36.2±2.53g/L)and S group(37.00±3.06g/L)(P<0.01).The N group and S group were simila(rP>0.05).④TheALT and creatinineof the rats in P group(45.10±5.13U/L)、(43.70±7.13μmol/L)were significantly higherthan the N group(33.30±4.32U/L)、(28.80±3.22μmol/L)and S group(32.90±2.99U/L)、(27.70±3.71μmol/L)(P<0.01).The N group and S group were similar(P>0.05).The urea、 uric acid and AST were similar among the differentgroups(P>0.05);⑤The serum level of NO、ET-1、TNF-α、IL-6in P group weresignificantly higher than the N group and S group(P<0.01),The N group and S groupwere similar(P>0.05).⑥The weight of fetus and placentas of the rats in P group(0.795±0.116g/0.353±0.056g)were significantly lower than the N group(1.097±0.197g/0.431±0.06g)and S group(1.155±0.211g/0.454±0.063g)(P<0.05),The N group and S group were similar(P>0.05).⑦The blood platelet of the rats inP group(1178±77*10^9/L)were significantly lower than the N group(1342±125*10^9/L)and S group(1361±203*10^9/L)(P<0.05),The N group and Sgroup were similar(P>0.05).Conclusion:①The STBM from preeclampsia women can inducepreeclampsia-like changes in pregnant rat.②The STBM from preeclampsia women cancause maternal vascular endothelial dysfunction and inflammation in vivo which maycontribute to the preeclampsia-like changes in rat。...
