Objectives1To investigate βⅢ-tubulin and microtubule destabilizing protein Stathmin (STMN)in thyroid papillary carcinoma (Papillary thyroid carcinoma, PTC) and in peritumoral tissues andin nodular goiter (Nodular goiter, NG) whether there are value of differential diagnosis of NGand PTC in expression and βⅢ-tubulin and STMN;2To study on the relationship betweenexpression of βⅢ-tubulin and STMN and clinical and pathological features of PTC;3Toexplore whether there is a relationship between the expression of βⅢ-tubulin and STMN in PTC.Methods Using immunohistochemistry technique in96cases of paraffin embedded specimens(21cases of peritumoral tissues as control group,25cases of NG,50cases of PTC chosen asexperimental group). According to the experimental results and Clinical medical records werestatistically analyzed by using the SPSS13.0software, P <0.05was statistically significant.Results1There16cases(68%)highly expressed of βⅢ-tubulin in PTC group, significantly higherthan that5cases(23.8%) in peritumoral tissue and9cases(36.0%)in the NG group, Fromthe peritumoral tissue to NG and then to PTC in the process of staining results showed enhancedtrend;2There17cases(68%)highly expressed of STMN in PTC group,significantly higher than that4cases(19.0%)in peritumoral tissue and11cases(44.0%)in the NG group,The same as βIII-tubulin from the peritumoral tissue to NG and then to PTC in the process of staining resultsshowed enhanced trend;3To combine the staining results of βⅢ-tubulin with clinicalpathological statistics in PTC group:the expression of βⅢ-tubulin in PTC is not related togender (P>0.05) and age (P>0.05),that the expression of βⅢ-tubulin in the cases with lymphnode metastasis is significantly higher than expression of the one without (χ2=4.112,P<0.05),it shows the expression of βⅢ-tubulin in PTC is related to lymph metastasis,that theexpression of βⅢ-tubulin in cases are in TNM III stage and IV stage is significantly higher thanexpression of the cases are TNM I stage and II stage (χ2=4.117,P<0.05),it shows it showsthe expression of βⅢ-tubulin in PTC is related to clinical pathological stage;4To combine thestaining results of STMN with clinical pathological statistics in PTC group:the expression ofSTMN in PTC is not related to gender(P>0.05) and age(P>0.05),that the expression ofSTMN in the cases with lymph node metastasis is significantly higher than expression of the onewithout (χ2=4.847,P<0.05),it shows the expression of STMN in PTC is related to lymphmetastasis,that the expression of STMN in cases are in TNM III stage and IV stage issignificantly higher than expression of the cases are TNM I stage and II stage (χ2=6.629,P<0.05),it shows it shows the expression of STMN in PTC is related to clinical pathologicalstage;5In34cases with high expression of βⅢ-tubulin there are27cases with high expression of STMN,STMN is lowly expressed in10cases out of13cases with low expression of βⅢ-tubulin,(χ2=8.517,P<0.05),βIII-tubulin and STMN are correlated with each other in PTC.βIII-tubulin and STMN are highly expressed in PTC with a statistical significance(P<0.05).There are more strong positive cases than the cases in NG group and control group(βIII-tubulin: P<0.05; STMN: P <0.05). The expression of βIII-tubulin and STMN both are irrelevant to age and gender (P>0.05), relevant to lymph node metastasis and clinical stage (P <0.05).Conclusions βIII-tubulin and STMN are highly expressed and correlated with each other inPTC,but lowly expressed in NG and peritumoral tissue. So βIII-tubulin and STMN may be usedfor differential diagnosis of PTC and NG. Though the expression of βIII-tubulin and STMN areirrelevant to age and gender and relevant to lymph node metastasis and clinical stage, so βIII-tubulin and STMN may be the molecular markers to predict the PTC process.
