Objective:To optimize the extraction technology, to establish the quality standard and the HPLCfingerprint of Xiangcaoguchi tincture. And then To make a systematic study of thepharmacy, making sure the controllability of quality.Methods:(1) Study on the extraction technology Using the contents of α-cyperone andwedelolactone in the extract solution and the yield of the dried extracts as the criteria,orthogonal test was used to optimize the conditions for the extraction process.(2) Study onQuality Standard Cyperus rotundus and Eclipta alba were qualitatively identified by thinlayer chromatography. The content of α-Cyperone and Wedelolactone were determined byhigh performance liquid chromatography.(3) HPLC fingerprint Analysis HPLC wasadopted in the fingerprint analysis, and the chromatographic conditions were as follows:PLATISIL ODS(250mm×4.6mm,5μm) column; mobile phase consisted ofacetonitrile-0.1%phosphorous acid solution(gradient elution); the detection wavelength at250nm; the column temperature of30℃; the flow rate of0.8mL·min-1.(4) The tentativestudy of stability To study the long-term stability and accelerated stability for six months,by investigating traits, differentiation, inspection and determination of content and so on.Results:(1) The best extraction conditions were as follows: The ethanol concentration was60﹪, thevelocity was5mL·min-1, and the immersion time was24h.(2) The TLC methord wasreproducible. The RSD in repeatability test of α-Cyperone was1.37%, the average recoveryrate was99.81%(RSD0.41%, n=6). The RSD in repeatability test of Wedelolactone was1.23%,the average recovery rate was100.16%(RSD0.8%, n=6).(3) There were mainly16 characteristic peaks in the fingerprints. Among these peaks,10peaks belonged to Cyperusrotundus L.,6peaks belonged to Herba Echiptae.(4) The indicators above were notchanged significantly, showing that the stability of the preparation was good.Conclusion:The extraction technology is stable and reasonable, and can be used for industrialproduction. The methord of quality standard and fingerprint is reliable, accurate andrepeatable, can be used to control the quality of Xiangcaoguchi tincture.
