Objective This study was to research the expression and significance of TLR4、NF-κB、IL-6、HSP60in peripheral blood of patients with hepatocellular carcinoma.Methods We set50patients with hepatocellular carcinoma for the research grop, then dividethem into AFP negative group and positive group by the level of serum AFP.30healthy volunteerswere randomly selected to be the control group at the same time. The expression of TLR4on thesurface of peripheral blood of mononuclear cell (PBMC) were tested by flow cytometry. Theexpression of NF-κB、IL-6、HSP60were tested by ELISA.Results The expression of TLR4on PBMC in patients withe hepatocellular carcinoma (HCC)were significantly higher than those in normal controls (P<0.01). The expression of HSP60andNF-κB、IL-6in patients with HCC were also significantly higher than those in normal controls(P<0.01). The expression of TLR4correlated with HSP60and NF-κB (r=0.343、r=0.352, P<0.01).The expression of TLR4、HSP60、NF-κB is much higher in AFP positive group compare with AFPnegative group (P<0.05).Conclusion TLR4may play an important role in the occurrence and development of HCC; Theexpression of TLR4is closely correlated with HSP60and NF-κB, they together promote thedevelopment of HCC; The expression of TLR4、HSP60、NF-κB is higher in AFP positive patientswith HCC, which reflect a heavier inflammation.
