Effect Of Echinococcus Granulosus Antigen B On Expression Of IL-2and IL-4in Type1Diabetes Gums

Objective:To study the effect of Echinococcus antigen B(EgAgB)on expression of IL-2and IL-4in gum tissue of STZ-induced T1DM murine model.For further research the role of EgAgB immune response mechanism in T1DM in the future, and to provide experimental evidence of immune therapy with T1DM in gingival inflammation.Methods:20female mice intraperitoneal injectioned of STZ by50mg/kg/d dose for5consecutive days for preparing T1DM mouse model, glucose values exceed16.7mmol/L and retest still do this levels the next day, suggesting that mouse model of T1DM successful modeling(20mice two times glucose monitoring all exceeded16.7mmol/L).20female mice divided into2groups(N=10),treatment group was established by intraperitoneal injection of100μg/10g EgB for5consecutive days, the control group received the same dose of normal saline.40weekend mice were sacrificed by cervical dislocation and whichever gums and total RNA was extracted with Trizol one-step,cDNA prepared by the reverse transcription(RT)reaction,use FQ-PCR to determine the variation of IL-2mRNA and IL-4mRNA gene’s expression between the treatment and control groups’gums.Result:Used SPSS17.0statistical package statistics analysis showed that:IL-2mRNA in EgAgB treatment group compared with the control group was significantly decreased(P<0.05); IL-4mRNA in EgAgB treated group than in the control group was significantly increased (P<0.05). Conclusion:EgAgB can make the ratio of cytokines produced by gum Th1/Th2cells change,it’s tend to Immune-related pathology Th2responses and affect the immune response mechanism of TIDM.
