Background: Currently, we pay close attention to the health ofpersons who were born during China famine years (1959-1961). The theoryof developmental origin for health and disease (DOHaD) has beenconfirmed by epidemiology. Meanwhile we found that early malnutritioncan affect cognition in adulthood, but the specific mechanism is unclear.Objective:To investigate the expression of phosphatidylinositol-3kinase catalytic subunit p110α in hippocampus of rats born with earlymalnutrition, and explore the possible mechanism of the early malnutritionon cognition of elderly rats.Methods:Fourteen-day pregnant rats were divided into control group(normal diet group) and experimental group (half amount of diet group).The newborn rats of the two groups were fed with normal diet after thepostnatal21days. The birth weight of rat pups was measured. Theperipheral insulin sensitivity of rats was evaluated by insulin tolerance test.The ability of learning and memory of offspring rats were assayed by Morris water maze test. The expression of PI3Kp110α in hippocampuswere detected by Western blot. The expression of Aβ1-42in cortex andhippocampus of offspring rats were detected by immunohistochemistry.Results:Compared with the normal control group, in experimentalgroup the mean birth weight of newborn rats was significantly lower(P<0.01), the peripheral insulin sensitivity were lowe(rP<0.05), the latencywas longer(P<0.05), the times of crossing platform was less(P<0.05),the percent time in quadrant was significantly shorter (P<0.01), theexpression of PI3Kp110α in hippocampus was lower (P<0.05) and thedifferent expression of Aβ1-42both in cortex and hippocampus were notstatistically significant (P>0.05).Conclusion:Early malnutrition impairs the cognition of elderly rats,which may be related to the abnormal expression of PI3Kp110α inhippocampus.
