Objective: In order to explore the role of macrophage and hepatocytein the occurrence of liver metaflammation and the potential mechanism.Methods: HepG2and THP-1differentiated macrophages were treatedwith different concentrations of PA (0mmol/L,0.08mmol/L,0.16mmol/L,0.32mmol/L) for24h, and cell proliferation was determined by MTT assay.Co-culture system of HepG2and macrophages was established by6-welltranswell-inserts(0.4μm pore size). The protein and mRNA expression ofinflammatory cytokines(IL-1β,IL-6,TNF-a) were evaluated by western-blotand real-time PCR, respectively. The CD163,Mannose-R and MCP-1geneexpression of macrophages were detected by real-time PCR. The migrationability of macrophages was measured by0.1%Crystal violet staining.Result: With the increasing concentrations of PA, there has nosignificant differences in cell proliferation in both HepG2and macrophages,and the inflammatory cytokines(IL-1β,IL-6,TNF-a) mRNA and proteinexpression of both HepG2and macrophages were highly improved, and theinflammatory cytokines mRNA expression of each cell type in the co-culture system was significantly elevated, compared with the controlgroup.The migration ability of macrophages was enhanced awfully, and thephenotype makers and MCP-1gene expression of macrophages werechanged in the co-culture system.Conclusion: These data indicate that PA could promote theinflammatory response in both HepG2and macrophages. Whenco-culturing HepG2with macrophages, the inflammatory response washighly exacerbated. The increasing migration ability of macrophages, andthe altering of the phenotype may make great contribution to theinflammation occurrence.
