Objective:By observing the speed and degree of rabbit radial fracture and the expression level of NT-3(neurotrophin-3) in the end of fracture in three groups of rabbits radial fracture models, to identify whether NT-3is usefull to fracture healing and provide the experiment of basis for orthopedic clinical application.Methods:Seventy-two New Zealand rabbits were choosed and radial fracture modles were established, randomly divided into three groups, group A (fracture+saline injection group), group B (fracture+NT-3injection group), group C (brain trauma with fracture+saline injection group). Each group was divided into4subgroups (n=6). Each group was injected vested drugs respectively at the lateral of thigh quadriceps at third and six days after operations. Each subgroup was selected from every group randomly, and detected the blood ALP (ALP) levels via ear source of venous every one week from one week to six week after operation,and kill them immediately after drawing blood. X-ray was performed at third and fourth week before animals sacrificed. The tissues of fracture end were got and hemateineosin(HE) stain was used to observe the osteotylus growth states and immunohistochemistry (IHC) stain was used to detected the expression of NT-3. The results were analysised by SPSS 17.0software.The results:1. X-ray findings:the degree of fracture healing was tested at the third week after operation, group C was best,then group B,group A the worst, and the comparison was more obvious in the fourth week.2. The expression of AKP:The contents of AKP were increased gradually after operation in the three groups. The growth trend of group A was gentle, however, group C and B were sharp.group B was more obvious.3. HE staining:Group A indacates the normal process of reactive hyperplasia and bone formation, group B was accelerated, and group C increased most significantly.4. NT-3IHC staining:The positive area ratio and OD value of NT-3in group A were steady after the operation. The peak values of group B and group C reached at the first and third week after operation respectively, then decreased gradually. Comparison among groups showed that the peak values were appeared at the first week in group B, and the difference was significent compared with group A and group C,while there was no difference (p>0.05) between group A and C. However,from the second week after operation, the differences were significent among pairwise comparison (p<0.05). The variation of mean value was group A<group B<group C. From the fourth week after operation, there was no difference (p>0.05) between group A and B. However, the difference (p<0.05) existed among other groups.Conclusion:NT-3can promote the process of rabbit radial fracture healing.
