Worldwide,lung cancer deaths account for about28%of the total,in the first to the causes of death from cancer.In our country,the incidence of lung cancer increased in11%annual rate.Due to the complexity of the biological characteristics of lung cancer and the limitations of modern medical treatment,comprehensive cancer treatment model has become an inevitable trend in the present and future lung cancer intervention,and the role of Chinese medicine in the comprehensive treatment of lung cancer causes more and more attention.Under the guidance of the theory of febrile diseases,to clear away heat and activate blood circulation method as the main treatment,self-made Jiawei Weijing decoction and apigenin are discussed systematically and deeply from the aspects of literature research,theory research and experiments.The first part:literature research.Elaborating the ancient traditional Chinese medicine understanding of lung cancer from the aspects of name,etiology,pathogenesis and therapeutic treatment.And then analyzes the current situation of traditional Chinese medicine treatment of lung cancer,and summarizes these main points:traditional Chinese medicine plays comprehensive function by improving the overall situation,inhibiting tumors growth and metastasis,modulating complications and cooperating with western medicine treatment.The second part:theory research.First of all,in the initial stage of lung cancer, healthy qi is damaged lightly yet, but inside the body there is a state of evil heat and solid content,as a result,phlegm,heat,and haemostasis will form tumor,so,at this moment,the treatment should be clearing heat and activating blood circulation.For another,there are three points:firstly,self-made Jiawei Weijing decoction clinical application and experimental research for the treatment of lung cancer,secondly,apigenin plays an important role of anti-tumor through various channels,thirdly,autophagy in the process of development and treatment of lung cancer is of great significance.The third part:experiment research.This study investigated the effects of Jiawei Weijing decoction and apigenin on lung adenocarcinoma A549cells,which can clear away heat and activate blood circulation,using modern molecular biology techniques from the perspective of autophagy regulation.The results show that the method has a good role in the inhibition of tumor cell growth,cell cycle arrest,apoptosis and autophagy-related signal transduction and so on.Growth inhibition:Containing serum of Jiawei Weijingtang and apigenin can inhibit the proliferation of A549cell,and showing a certain dose-effect and time-effect relationship.Apoptosis:Under the fluorescence microscope,shedding cells and apoptotic bodies show that both drugs can induce the apoptosis of A549lung cancer cells.Self-made Jiawei Weijing decoction and apigenin can induce apoptosis inhibition of lung cancer by the proliferation of A549cells and then play a role of treatment.according to more shedding cells,more apoptotic bodies and higher apoptosis rate, apigenin inhibits the growth of A549cells mainly through way to induce apoptosis.Cell cycle arrest:the two drugs both have an effect on A549cell cycle.compared with blank control group,the cycle distribution has significant change (P<0.05).But the specific influence of related parameters is different obviously:after the same time,apigenin of50μmol/L is significantly higher than the10%containing serum on the inhibitory effect(P<0.01),and within the scope of the concentration gradient of the experimental setup,apigenin element on A549cell proliferation inhibition is superior to the containing serum of self-made Jiawei Weijing decoction.In terms of cycle arrest,containing serum of self-made Jiawei Weijing decoction exert anti-tumor effect by arresting A549cells in G2stage and preventing cell mitosis. Apigenin can also arrest A549cells in G2stage but not presented dose-effect and time-effect relationship.Signaling pathway:both containing serum of self-made Jiawei Weijing decoction and apigenin can inhibit the high expression of mTOR、PI-3K protein (P＜0.01)In this study,preliminary clarification about self-made Jiawei Weijing decoction and apigenin resistance mechanisms of lung carcinoma A549on the cellular and molecular level,had an in-depth discussion on the main link of lung cancer treated by Chinese medicine,so as to provide reliable laboratory evidence of clinical treatment for early lung cancer.
