Objective:To observe the growth of the osteotylus of type2diabetes rats and probe into the relationship between the change of expression of Osx、Dlx5in callus of type2diabetes rats and fracture healing disorder.Methods:32male SD rats with proper weight in eight weeks age are divided into control (n=14) and experimental (n=18) groups,The experimental group is fed with high fat and sugar diet,The contrast group is fed with normal diet,After eight weeks, taking the blood from rat’s tails.The experimental group is intraperitoneal injected with STZ (30mg/kg), while the control group receive intraperitoneal injection of the same amount of citrate buffer.Two more weeks later,we select the rats whose blood glucose is above16.7mmol/L in the control group to continue with the next experiments.After establishing left tibia traction fracture model in the two groups,beginning to extend the external fixator in the first postoperative day,the extension of length is0.15mm each time, twice a day. After2weeks, the rats are executed,then blood samples and the left tibia specimens are collected. The amount of new bone callus area, the number of fat cells per unit area will be measured by radiologic and histological methods, the expression of Osx, Dlx5mRNA will be measured by QPCR.Results:After8weeks of raising, TC was significantly higher in the experimental group than that in the contrast group (P<0.01),compared with the control group,TG was increased (P<0.05),FBG and FINs which compared with the control group made no difference (P>0.05),Experiment10weeks and at the end of the test,the experimental group of rats TG,TC,FBG increased compared with the control group (P <0.05) while FINs had no statistical significance (P>0.05).X-ray:The osteotylus of Experimental group than the control group significantly reduced,HE staining showed the micro column bone (MCF) had shorten and disordered arrangement in the experimental group which was compared with control group and most shallow dye in the initial matrix forefront.The area of new born osteotylus in The experimental group was significantly reduced,while the number of fat cells per unit area was increased (P<0.01).QPCR detection is displayed:the experimental group than the control group Osx osteotylus tissue expression was decreased (P <0.05), Dlx5expression was decreased (P<0.05).Conclusion:In comparison with the normal rats,type2diabetes rats had a longer time in fracture healing. The expression of Osx and Dlx5are decreased in bone tissue,which may be one of the barrier to fracture healing of type2diabetes.22pictures,13tables,28references.
