| Objective To explore the effect and mechanism of BSHXF on promoting fracture healing and evaluate the curative effect via varies testing measurements,including X-ray,Micro-CT,Histology,Histomorphometry,qPCR and so on.Methed The C57BL/6 mouse was choose in this experiment,all of them were operated a unilateral open tibiae transverse fracture with intramedullary needle fixation model.After the operation.X-ray was used to sure a qualified fracture model was established,then the qualified were divided into 2 groups 5 time points randomly.The BSHXF and control group were treated with BSHXF and saline respectively.The characteristics of bone fracture healing of different groups were serially analyzed and compared by X-ray,Micro-CT,Histology,Histomorphometry,qPCR.The observation of body weights,mobility were included.Time points were set by day 4,7,10,14,21 postoperatively.Result 1.X-ray results show blurry degree of fracture lines at day 14 and 21 postoperatively in mice treated with BSHXF was significantly higher(P<0.05).2.Callus mineralized volume fraction of BSHXF group was significantly higher in comparison to control group by day7,10,14 postoperatively(P<0.05).3.Callus bone volume was significantly higher compared to control group by day 7,10,14 postoperatively(P<0.05).4.The Mg.Ar/Ps.Cl.Ar in the BSHXF group was significantly higher than that in the control group(P<0.05)in the 7th,10th,14th and 21st days after surgery.5.The Cg.Ar/Ps.Cl.Ar in the BSHXF group was significantly higher than that in the control group(P<0.05)in the 7th day after surgery.6.The recovery of weight and activity of mice in the BSHXF group was faster than that of the control group.7.The expression of Sox9 gene and Col2al gene in mice in the BSHXF group was concentrated in the 7th and 10th days after surgery,with statistical difference(p<0.05).The expression level of CollOal was significantly higher in the 7th and 10th days day after surgery,with statistical difference(p<0.05).The expression level of Runx2 was concentrated in the 10th day after the operation,and the BSHXF group was significantly higher than the control group(p<0.05).The expression levels of Osteocalcin gene in the BSHXF group were higher than that in the control group in the 7th,10th,14th and 21st days.The expression of Osteocalcin gene in the 10th and 14th days was significantly higher,with a statistically significant difference(p<0.05).The expression level of Tgfbr2 gene in the BSHXF group was significantly higher than that of the control group in the 7th,10th and 14th days after the operation(p<0.05).8.Then protein expression of pSmad2 was increased at day 4 in the BSHXF group(P<0.05).Conclusion 1.The BSHXF can promotes tibiae fracture healing of mouse.2.The BSHXF maybe upregulate pSmad2 and activeTGF-βsignaling pathway to promote fracture healing. |
