| Objective:A preliminary understanding of its biological tissue compatibility and security was obtained by vivo and vitro animal experiments of nano-scale freshwater pearl powder with milling preparation, in order to provide reference data for further appplying research in experimental bone repair materials.Methods:The nano-scale freshwater pearl powder was prepared by mechanical ball milling method (Vertical biaxial nano-ceramic sand grinding dispersion machine, micron-scale freshwater pearl powder and ethanol in the ratio of weight to volume ratio of about1:15). The nano-scale and micron-sized freshwater pearl freshwater pearl powder was analyzed and compared by Laser particle size analyzer (LPSA), Scanning electron microscope (SEM), X-ray diffraction (XRD), Transmission electron microscopy (TEM), Kjeldahl method and reference Determination of amino acids in foods (GB/T5009.124-2003) etc. Reference to the internationally recognized (GB/T)16886/ISO10993(Biology evaluation of medical devices) series of standards, the cytotoxicity test, hemolysis test, intracutaneous stimulation test and acute systemic toxicity test on nano-scale freshwater pearl powder were applied to study the biocompatibility and biosafety preliminarily.Results:(1) Nano-scale pearl powder could be prepared by nanometer ceramic sand grinding dispersion machine Which is a set of integrated dispersion and efficient ball mill grinding.(2) Scanning electron microscopy, X-ray diffraction, transmission electron microscopy, Laser particle size analyzer can detect nanomaterial’s particle size, while the particle size detection of nano-scale pearl powder requires a combination of different detection methods to comprehensive analysis due to the different detection principles and extremely easily agglomeration of nanoparticles.(3) Absolute alcohol as a liquid medium, the protein and amino acid content of nano-scale freshwater pearl powder prepared by ball milling did not changed significantly.(4) Cytotoxicity test:the nano-scale pearl powder-DMEM extract cytotoxicity was I level, in line with the ISO standards of cytotoxicity.(5) Hemolysis test:the nano-scale pearl powder-saline extract hemolysis rate was1.01%, less than5%, in line with ISO standards of hemolysis.(6) Intracutaneous stimulation test:the intracutaneous stimulation index of nanos-cale pearl powder-saline extract was0, in line with ISO standards of intracutaneous stimulation.(7) Acute systemic toxicity test:in the experimental observation period, the experimental group and the control group SD rats have no toxic symptoms, no adverse reactions, no death, and no weight gain abnormal. One week later, the examination of abdominal cavity and the histopathology of liver, kidney and spleen are normal.Conclusions:With absolute ethanol as the medium, micron pearl powder can be ground into nano-scale by wet ball milling method, The protein and amino acid content of the nano-scale pearl powder did not change significantly.
