Objective:More and more experimental evidence that hypertension is a chronic inflammatory disease, the activation of lymphocytes (especially T lymphocytes) participating in the development of hypertension, the potassium channel of lymphocytes is crucial for lymphocyte’s activation. To study the effects of the ARJB drugs telmisartan on the potassium channels of T-lymphocyte in peripheral blood of patients with hypertension in Xinjiang Kazakh and provide some experimental evidence for the new therapeutic targets of hypertension. Methods:30Xinjiang Kazakh hypertensive patients without antihypertensive therapy were randomly selected from hypertensive clinic service at the First Affiliated Hospital of Xinjiang Medical University from June2012to December2013, and the patients were randomly divided into blank control group, telmisartan group and potassium channel blockers (positive control) group, there were10patients in each group(5men and5women). Peripheral blood T lymphocytes which were isolated by magnetic cell sorting were cultured in vitro to make them activation and proliferation, and Continue to foster Oh,24h,48h respectively after given the appropriate packet processing. Using the patch-clamp technique to detect the electrophysiological properties changes of Kv1.3and KCa3.1potassium channel after activation and proliferation of the T lymphocytes, and using the technology of real time quantitative PCR(Real-time PCR) and western blotting to detect the Kvl.3and KCa3.1channels mRNA and protein expression changes after activation and proliferation of the T lymphocytes. Results:With the extension of the intervention time, telmisartan can effectively block the activation and proliferation of T lymphocytes Kv1.3and KCa3.1potassium channels currents, and can effectively inhibit the mRNA and protein expression of these channels.(1) The whole-cell currents, mRNA and protein expression of the Kv1.3and KCa3.1channels after the activation and proliferation of T lymphocytes when telmisartan intervene in24h and48h were significantly reduced compared with the control group (all P<0.05);(2) There were significant differences of the whole cell currents, mRNA and protein expression of the Kvl.3and KCa3.1channels after the activation and proliferation of T lymphocytes at different time point in telmisartan group(all P<0.05). Conclusion:Telmisartan can effectively block the potassium channels of T lymphocytes in patients with hypertension, suggesting that telmisartan has anti-inflammatory effect of hypertension.
