| Objective:To study the effects of telmisartan on NLRP3 inflammasomes signaling pathways related molecular expression of T-lymphocyte in peripheral blood of patients with Hypertension in Xinjiang Kazakh, and explore to the pathogenesis of hypertension and provide experimental basis for new targets for treatment.Methods:From 2013.07 to 2014.03 in Xinjiang medical university first affiliated hospital heart center outpatient visits for the first time selected randomly and without antihypertensive drug treatment of kazak in Xinjiang (male-female) of 30 cases of patients with high blood pressure, will be randomly divided into blank control group, telmisartan group and potassium channel blockers 3 groups, each group of 10 cases, gender parity.Take immune magnetic beads to separate groups of patients with peripheral blood T lymphocyte proliferation in vitro activation, give corresponding continue to cultivate Oh, 24h,48h after.Using the technology of real time quantitative PCR(Real-time PCR) and western blotting to detect the NLRP3 and Caspase-1 mRNA and protein expressions changes after activation of the T lymphocytes.Results:No significance of general condition between three groups was after intervention of telmisartane with the extension of the intervention time, the mRNA and protein expressions of NLRP3 and Caspase-1 in T lymphocytes were decreased. (1) The whole-cell currents, mRNA and protein expression of the NLRP3 and Caspase-1 after the activation and proliferation of T lymphocytes when telmisartan intervene in 24h and 48h were significantly reduced compared with the control group (all P<0.05); (2) There were significant differences of the whole cell currents, mRNA and protein expression of the NLRP3 and Caspase-1 after the activation and proliferation of T lymphocytes at different time point in telmisartan group (all P<0.05).Conclusion:Telmisartan can effectively restrain high blood pressure patients peripheral blood T lymphocyte NLRP3 inflammatory corpuscle signaling pathways related to molecular expression, prompt telmisartan has inflammation of antihypertensive effect, provide new evidence for high blood pressure, inflammation theory, at the same time for the anti-inflammatory treatment of high blood pressure to find new research direction. |
