Objective:This essay makes a comprehensive comparative study of Bulbus Fritillariae pallidiflorae from different habitats and different wild growing and cultivated species,of which provide basis for cultivated planting of Bulbus Fritillaria pallidiflorae can been substituted to clinic in Traditional Chinese Medicine.Methods:1) According to the2010edition of Pharmacopoeia,comparative study traits,micro powder,TLC identification, moisture, total ash and extract for Bulbus Fritillariae pallidiflorae from different wild growing and cultivated species;2) As a supplement to improve Pharmacopoeia data, the microstructure characteristics of wild and cultivated Fritillaria, heavy metal preliminary examination and UV-VIS method for the determination of total alkaloid content were studied;At the same time, by means of modern analysis technology, content by using the UPLC-ELSD method, HPLC method of wild and cultivated Fritillaria alkaloids and Sipeimine-3β-D-glucoside and sipeimine, water-soluble components β-thymidine and adenosine were determined,In different places of wild and cultivated Fritillaria alkaloids and water-soluble constituents of fingerprint, and uses the Pharmacopoeia Commission promulgated the "chromatographic fingerprint similarity evaluation system for2004A edition" software was used to establish fingerprint common pattern, calculate the similarity graph spectrum, analysis results using cluster analysis method and principal component.Results:1) The traits,micro powder, TLC identification, moisture, total ash and extract for Fritillariae pallidiflorae from different wild growing and cultivated species were consistent with the2010Edition Pharmacopoeia,in the character of the artificial cultivation of Fritillaria pallidiflora in general than the wild products to high, big, texture than the wild product is loose, no significant differences in the rest hand.2) Microscopic structure of wild and cultivated Fritillaria pallidiflora consists of epidermis, cortex parenchyma, limited external phloem parenchyma cells of vascular bundles,Parenchyma cells are cultivated wild Fritillaria arranged a little closer together,The number of vascular bundle of Fritillaria walujewii Regel are slightly larger than Fritillaria pallidiflora Schrenk.Cultivation of different origin of Fritillaria pallidiflora heavy metals shows that heavy metals and harmful elements of Fritillaria pallidiflora cultivated products meet the national requirements of pharmacopoeia;Measured the levels of different areas of wild and cultivated Fritillaria alkaloid content and water soluble components of β-thymidine and adenosine larger;The total content of Sipeimine-3β-D-glucoside and sipeimine reached total Pharmacopoeia requirements shall not be less than0.070%;Different habitats of wild and cultivated the29batch of Fritillaria pallidiflora alkaloids fingerprint identified16common characteristic peaks, water-soluble components fingerprint identified14common characteristic peaks,UPLC-ELSD alkaloids HPLC fingerprints of water-soluble fingerprint common pattern is established respectively.Conclusion:The traits,micro powder, TLC identification, moisture, total ash and extract for Fritillariae pallidiflorae were consistent with the2010Edition Pharmacopoeia;To improve the Pharmacopoeia supplement data: microstructure, heavy metals arid water soluble components of β-thymidine and adenosine content method;Methods UPLC-ELSD fingerprints and HPLC fingerprints of the fast, with precision, repeatability, stability is good, which provide basis for cultivated planting of Bulbus Fritillaria pallidiflorae can been used to clinic in Traditional Chinese Medicine.
