ObjectiveThe intention was to probe the effect of human retinal vascular endothelial cellproliferation and its VEGF expression on hedyotis diffusa injection and its angiogene-sis inhibition mechanism in high glucose environment,and to provide a new treatme-nt for Diabetic retinopathy.MethodsF Ⅷ r-Ab was identified HRCECs which were cultured in vitro.The culturedHRCECs were dealed with different concentration of hedyotis diffusa injection,theinfluence of cell proliferation was surveyed by CCK-8.The VEGF expression ofHRCECs was identified by using ICC(Immunocytochemistry) test and ELISA(Enzy-melinked immunosorbent assay).Results1.ICC assay indicated that in HRCECs the expression of F Ⅷ r-Ag was positive.2.CCK-8assay indicated that with6.25ml/L、12.5ml/L、25ml/L、50ml/L HDI treatmentunder high glucose,it inhabited the ability of HRCECs proliferation dependanting ontime and dose,except6.25ml/L HDI group.The inhibitory rates were3.58±1.06%、8.92±3.32%、15.68±3.18%、33.17±2.49%after effected by HDI on24hours;they are4.17±1.53%、14.09±2.98%、31.40±3.71%、45.95±2.63%after48hours and4.53±1.09%、24.18±6.53%、47.62±2.96%、63.14±3.37%after72hours;the differencebetween HDI treatment groups and high glucose control group was significant(P<0.05),as soon as the difference between the high glucose and the low glucosecontrol group(P<0.05).3. ICC test and ELISA assay both showed that with12.5ml/L、25ml/L、50ml/L HDI treatment, after72hours,the difference of the picture with thegray value and OD value was statistics significant (P<0.05), the same as thedifference between the high glucose and the low glucose control group (P<0.05). ConclusionHDW inhibited the proliferation HRCECs dependenting on dose and time;it caninhibit angiogenesis by decreasing the VEGF expression. It has important applicationsin the treatment of retinal neovascularization in DR.
