Objective: A urosepsis model is established in the rabbit. To investigate The functionof exogenous hydrogen sulfide on cytokines in kidney tissue of urosepsis rabbit modeland the mechanism it works, then the expression of cytokines will be tested in renaltissue of the rabbit model by hydrogen sulfide treatment.Method:1、To established Acute urinary tract obstruction and infection induced urosepsismodel in the rabbit,colibacillus was injected in the left ureter of the model and ligatethe ureter.2、 The48healthy male rabbit were assigned to control group,sham-operatedgroup,sepsis group and NaHS group at random,12rabbits each group.3、Anal temperature, respiratory rate and heart rate in12h,24h,36h and48h afteroperation was detected.4、Peripheral blood cell counts and renal function tests were completed12h,24h,36hand48h after operation.5、The renal tissue of rabbit of different points in time was stained by HE andmorphological change was observed. Expression of TNF-α、IL-10in renal wasdetected by Western blot,and gene level of NF-κB was detected by reversetranslation-PCR.Result:1、Vital Signs:Compared with control group,temperature, respirations and heart rate ofsham-operated group had no significant difference(p>0.05). With the extendedresponse time, the values of vital signs of sepsis group were on the rise and higherthan control group and sham-operated group(P<0.05), the values of NaHS groupwere slightly below sepsis group(P<0.05).2、Blood index tests:compared Peripheral blood cell counts and renal function testsbefore operation in each group,the difference have no statistics significant.Compared with sham-operated group,peripheral blood cell counts and renal function tests in12h,24h,36h and48h after operation of control group had no significantdifference(p>0.05). With the extended response time, the values of sepsis group wereon the rise and higher than control group and sham-operated group obviously(P<0.05), the values of NaHS group were slightly below sepsis group(P<0.05).3、Pathomorphology:under light microscope, glomerulus atrophy, Renal capsuledeformation, the tubular revealed dilatation and inflammatory cellular infiltration inrenal inerestitum can also be observed.Compared with sepsis group, the extent ofchange of NaHS group were lighter.4、Western Blot test:Compared with sham-operated group,the expression of TNF-αand IL-10in renal tissue of control group has no significant difference(P>0.05). Withthe extended response time, the expression of sepsis group were on the rise and higherthan control group and sham-operated group obviously(P<0.05), the values of TNF-αof NaHS group were slightly below sepsis group when IL-10were even higher(P<0.05).5、RT-PCR test:Compared with sham-operated group,the expression of NF-κB ofcontrol group has no significant difference(P>0.05). With the extended response time,the expression of sepsis group were on the rise and higher than control group andsham-operated group obviously(P<0.05), the values of NaHS group were slightlybelow sepsis group(P<0.05).Conclusion:1、The urosepsis model can be established succefully.2、exogenous hydrogen can protect kidney function of urosepsis by downregulatingNF-κB and TNF-α,and upregulating IL-10at the same time.
