Objective:To investigate the effect of ulinastatin on urosepsis induced by lipopolysaccharide which was injected into the ureter ligated, so as to provide theoretical and practical basis for theraping urosepsis.Method:1.To make the model of urosepsis in rabbits by the infusion of lipopolysaccharide to the left ureter ligated.2.Twenty-four healthy male rabbits were randomly divided into four groups(N=6each). normal control group; sham operation group; model group and ulinastatin group.3. Rectal temperature(T), respiration rate(RR) were examed:Peripheral blood cell counts(WBS, PLT), serum of C reaction protein(CRP) were tested after36hours of the experiment; The levels of TNF-α and IL-10in serum of each group were measured by ELISA; The liver, kidney and lung specimens were harvested for HE stain to observe modality changes of each tissue; Immunohistochemistry(IHC) method was used to detect TNF-α and IL-10in renal tissues of the experimental rabbits.Result:1. The general status of ribbitsThe T and RR between sham group and normal group didn’t have significant statistical differences(P>0.05); The T and RR in model group was significantly higher than normal group and sham group(P<0.05) and UTI group was lower than model group (P<0.05).2. Bloodtest index results The WBC counts between sham group and normal group didn’t have significant statistical differences(P>0.05); The WBC of blood in model group was significantly higher than that in normal group and sham group(P<0.05). Compared with model group, the UTI group was decreased(P<0.05).The PLT counts shows that there were no significant statistically differences in each group(P>0.05).Serum of C reaction protein(CRP) results:Compared with normal group, the serum of CRP in sham group was increased, there was significant statistical differences(P<0.05); model group was significantly higher than that in normal group and sham group(P<0.05). Compared with model group, the UTI group was significantly decreased(P<0.05).Serum of TNF-α, IL-10resuls:Compared with that of normal group, the serum of TNF-a, IL-10in sham group didn’t have significant statistical differences(P>0.05), the serum of TNF-α in model group was significantly higher than sham group(P<0.05); Compared with model group, the serum of TNF-a in UTI group was significantly decreased(P<0.05), the serum of IL-10was opposite with TNF-a.3. HE dyeing resultsThe liver, kidney and lungs were normal in normal group and sham group; In model group, hepatocyte little turgid swelling and cytolymph relaxation, inflammatory infiltrate; renal glomerulus deformities and renal tubules obvious expansion, renal interstitium were seen inflammatory infiltrate;pulmonary alveolar septum diffuse increased thickness and inflammatory infiltrate. Compare with than in model group, UTI group was slightly lighten on all inflammatory reactions.4. TNF-α, IL-10protein IHC resultsTNF-α, IL-10staining was noted in normal group and sham group; The TNF-a protein expression was enhaneed in model group, while UTI group had lower expression than that in model group; The IL-10protein expression was opposite with TNF-a.Conclusion: 1. We infuse the ureter with lipopolysaccharide, then ligate it for36hours, so the urosepsis animal models can established successfully.2. The experimental result found that UTI can effectively control sepsis and relieve inflammatory reaction of tissues and organ.The possible mechanism of UTI effectively control sepsis is it can effectively prevent and control sepsis events by down-regulate TNF-a, and up-regulate IL-10.
