【Objective】To investigate the expression differences of TUBA1A gene in bone marrows ofpatients with different hematologic malignancies, analyze the expressioncharacteristics and significances in hematologic malignancies.【Methods】1. Collected79cases of five kinds of different bone marrow samples of patientswith hematologic malignancies (Acute lymphoblastic leukemia ALL16cases, Acutemyeloid leukemia AML18cases, Chronic lymphocytic leukemia CLL10cases,Chronic myeloid leukemia-Chronic phase CML-CP15case, Multiple myeloma MM20cases)。While collecting20samples of light-moderate iron deficiency anemia(IDA) patients’s bone marrow2ml as a control group, rule out other malignanciescombined;2. Separating mononuclear cells with lymphocyte separation medium;3. Immunoblotting (Western Blot) to detect TUBA1A protein level in fivekinds of hematologic malignancies patients’ bone marrow cells and in the controlgroup, and analyze statistically;4. Reverse transcription-polymerase chain reaction (RT-PCR) to detectTUBA1A mRNA level in five kinds of hematologic malignancies patients’ bonemarrow cells and in the control group, and analyze statistically;5. Fluorescence quantitative realtime polymerase chain reaction (qRT-PCR) toamplify bone marrows’ cDNA of five patients with hematologic malignancies,analysed TUBA1A expression levels.【Results】1. Five kinds of bone marrow of patients with hematologic malignancies andpatients in the control group all have TUBA1A expression.2. TUBA1A mRNA and protein expression levels are higher in bone marrow cells of patients with ALL、AML、MM than the control group’s, The difference wasstatistically significant (P<0.05), however TUBA1A mRNA and protein expressionlevels have no significant difference in bone marrow cells of patients with CLL、CML-CP than the control group’s (P>0.05)。3. TUBA1A expression levels are different in bone marrow cells of patients withdifferent hematologic malignancies, with the highest expression in multiple myeloma(MM) patients.【Conclusion】1. TUBA1A expressed differentially in five kinds of hematologic malignancies,It has a wide range of expression in bone marrows of patients with acute leukemia andmultiple myeloma, with the highest expression in multiple myeloma.2. The expressions have no significant differences in the expression in bonemarrows of patients with Chronic myeloid leukemia-chronic phase, Chroniclymphocytic leukemia and control group. sample volume needs to be expanded forfurther research.
