Objective: To compare the patients with hematologic malignancies andhealthy circulating DNA levels, and monitoring the change of circulating DNAlevels before and after chemotherapy in patients with hematologic malignancies.Methods: Picogreen fluorescent dye was applied to80patients withhematologic malignancies and circulating DNA quantitative analysis of20cases of healthy people.Results:①Circulating DNA electrophoresis bands was between100~1000bp.②循环DNA levels of patients of Acute leukemia without chemotherapy were123.756±74.558ng/ml, circulating DNA levels of patients of lymphoma withoutchemotherapy were8.942±3.900ng/ml, both of them were higher than the循环DNA levels of healthy people (2.008±0.869ng/ml),P values <0.05, withstatistical significance.③Circulating DNA levels of Acute leukemia withoutchemotherapy (123.756±74.558ng/ml) were higher than lymphoma withoutchemotherapy (8.942±3.900ng/ml),P values <0.05, with statisticalsignificance.④Circulating DNA levels of Acute Leukemia after chemotherapy(26.007±18.403ng/ml) were less than without chemotherapy (123.756±74.558ng/ml), a73.50%drop, circulating DNA levels of Lymphoma afterchemotherapy (2.370±1.108ng/ml) were less than without chemotherapy(8.942±3.900ng/ml), a78.99%drop.⑤circulating DNA levels of Lymphomaafter chemotherapy were close to the levels of healthy people, circulating DNAlevels of Acute Leukemia after chemotherapy were still higher than the levels ofhealthy people.⑥Compare to patients with poor prognosis, circulating DNAlevels of patients with good prognosis were more significantly decreased. Conclusion:①Circulating DNA levels had significant difference betweenmalignancies patients and healthy people. Circulating DNA levels of acuteleukemia were higher than lymphoma, with significant difference.②CirculatingDNA levels of malignancies patients declined after chemotherapy significantly,lymphoma patients could decrease close to the levels of healthy people.③Thedetection of levels of circulating DNA makes sense in prognosis ofmalignancies.
