Objective:Through observing the "Zusanli" electro-acupuncture(EA) preconditioning protective effects of triptolide induced acute liver toxicity in rats, to provide experimental basis for acupuncture attenuated.Methods:25SD rats were randomly divided into control group, EA group and medicine group, blank group5, acupuncture group and medicine group with10rats in each group. EA group rats in electro-acupuncture "Zusanli" stimulus processing, frequency5Hz, the intensity of4mA,1times a day, treatment for5consecutive days. Then in the reference basis, the triptolide standard at a dose of3.0mg/kg propanediol fully dissolved and diluted with saline into stomach perfusion solution, the control group, EA group were administered to the rats in blank group, administered by propylene glycol and saline equal to service, drug group and acupuncture group two groups of rats acute liver toxicity of triptolide model, to observe the behavior of rats and the number of deaths. All rats were killed after24hours, the left hepatic lobe at the same site tissue sections, liver pathologic changes were observed in hepatic tissue; at the same time, take a piece of liver tissue at the liver maximum leaf, weighing, measuring the liver tissue homogenate, alanine aminotransferase (ALT), alanine aminotransferase (AST), malondialdehyde (MDA), superoxide dismutase enzyme (SOD), nitric oxide (NO), tumor necrosis factor alpha (TNF-α), interleukin10(IL-10) content. Groups were compared using analysis of variance, the comparison between two two groups was performed using LSD test, P<0.05was considered statistically significant.Results:The liver tissue AST, ALT, MDA, TNF-a, NO content EA group than the medicine group (P<0.05), but still higher than the control group (P<0.05); SOD content EA group than the medicine group (P<0.05), and the difference compared with the control group no statistical significance (P>0.05). IL-10levels there is an upward trend in the EA group compared to the medicine group, but the difference was not statistically significant (P>0.05). Compared with the control group, EA group IL-10content was reduced (P=0.05)Conclusions:Rats Established rats acute liver injury model.by feeding the Triptolide, The dose of3.0mg/kg, observed24h hours is relatively appropriate dosesand relatively suitable time, Electroacupuncture Pretreatment has certain protective effects on rat liver cells. But this protective effect is relative, Do not completely inhibit the acute liver injury caused by Triptolide. Electroacupuncture pretreatment can improve the antioxidant potential. Keep SOD at a high level, inhibit the production of MDA, Improve rats tolerance of Triptolide. Inhibits TNF-α, NO proinflammatory factor, But less effect against inflammatory cell factor IL-10.
