[Abstract] Background:Acupuncture at Neiguan could improve cardiac function and neovascularization, cardiomyocyte survival, and inhibit cardiomyocyte apoptosis. But its mechanisms are not yet fully uncovered. Aim:Our study demonstrates that acupuncture can effectively up-regulate VEGF expression through H3K9acetylation modification directly at the VEGF promoter and hence activate VEGF-induced angiogenesis in rat MI models. Methods:60SD male rats were devide into4groups: Control group;Control-EA group(Control+Neiguan); Model group (MI at LAD); EA group (MI+Neiguan). Result:1. The VEGF and H3K9ace expression of MI group was decreased compared with Control groups (P<0.05);2. EA treatment strikingly increased the VEGF and H3K9ace expression compared with model groups (P<0.05);3.The VEGF and H3K9ace expression of EA treatment group has no significant difference compared with Control groups;4. The VEGF and H3K9ace protein expression of Control-EA group was decreased compared with EA groups (P<0.05);5. The VEGF and H3K9ace expression of MI group has no significant difference compared with Control-EA groups;6. The VEGF and H3K9ace protein expression in Control groups significantly increased compared with Control-EA groups (P<0.05).;Conclusion:Acupuncture at Neiguan could improve the VEGF gene expression suggesting a possible mechanism by which acupuncture improves cardiac function when blood supply is reduced by ischemic injury.
