Background:In recent years,the research of ischemic cardiomyopathy has progressively moved into the molecular and gene level.A large number of experiments have proved that acupuncture has effect on improving myocardial ischemia.A large number of research results of basic animal experiments and clinical studies have been generated by gene chip and next generation sequencing.However,limited by various factors,there is a great difference between the clinical studies and the basic animal experiments.No research has been done on the effective comparison and analysis of animal and clinical experiments so far,and no one has put forward the data processing scheme of the combination of them.This is a huge obstacle to the mutual promotion of clinical and experimental studies.Objective:Compare the similarities and differences of the gene profiles,the key genes and pathways under myocardial ischemic conditions in different experimental subjects after acupuncture treatment.Smaples were harvested from neutrophils of peripheral blood of patients or ischemic heart tissues of rats.Data were analyzed by bioinformatics.The results care expected to be proposed for clinical experimental research and animal models in the use of animal models for human disease research and provide guidance for feasibility.Methods:Based on the original data of high throughput sequencing of different subjects in our group,we screened by bioinformatics method,screened from log2(fold-change,FC)of gene expression difference multiplier(criteria of log2(FC)?|±1|,>1 is up-regulated,<-1 is down-regulated),to search for CO regulated genes.Using DAVID to annotate gene function,and on the basis of KEGG_PATHWAY pathway,use String to analyze and predict protein protein interaction network,filter and evaluate the data of functional genomics,and provide the prediction of species across species.Conclusion:Our study found that both clinical and animal experiments have improved in the following pathways:mitochondria,energy metabolism and redox reaction,homeostasis,inflammation,myocardial contraction,and TNNT2,ACTC1,ATP2A2,TNNC1,MYH6,TPM1,which play an important role in improving the protection against myocardial ischemia in acupuncture PC6.However,the significant changes of CXCR2 and IL1R2 genes in clinical experiments were not found in animal experiments.Only the related genes related to XCL1,CCR10,CXCL,CCR10 and other related genes were changed obviously,indicating the difference between the clinical and animal experiments due to the models and materials.Some conclusions may not be shared in clinical and animal experiments,and theresults of experimental studies in small mammals cannot be directly used in large mammals.Some current experimental models need to be improved to mimic human disease model better.
