ObjectiveTo observe the effects of Yiqi-huayu-huatan-yangyin preparation onthe liver histopathology and the expression of FSP1、PAR-2mRNA andprotien in liver fibrosis rats induced by CCl4.Methods1.Experimental groups: The SD rats were randomly dividedinto:normal group,model group,colchicines group and low/moderate/highdosages of Yiqi-huayu-huatan-yangyin preparation group.Rats in thenormal and model groups were fed with normal saline,rats in colchicinegroup were fed with0.1mg/kg·d colchicine and rats in low/moderate/highdosages of Yiqi-huayu-huatan-yangyin preparation group were fed withYiqi-huayu-huatan-yangyin(30,60,120g/kg·d)correspondingly.2.The pathological changes of the liver tissue were observed by bothlight and electron microscopy.3.Activities of ALT and AST in liver tissues were evaluated byspectrophotometry.Levels of HA and CⅣ in serum were determined usingrat ELISA kit.4.The expression of FSP1、PAR-2mRNA in the liver tissue weredetected by qRT-PCR. 5.The expression of FSP1、PAR-2protein in the liver tissue weredetected by Western blotting.Results1.Levels of ALT,AST,HA and CⅣ in serum、fibrosis changes and theexpression of FSP1、PAR-2mRNA and protein of liver tissues wereincreased significantly in model group compared with those in normalgroup(P<0.05).2.In contrast with the model group,levels of ALT,AST,HA and CⅣ inserum、fibrosis changes and the expression of FSP1、PAR-2mRNA andprotein of liver tissues were decreased obviously in treatmentgroups(P<0.05).3.Among all the treatment groups,the anti-fibrotic effects in moderateand high dosage of Yiqi-Huayu-Huatan-Yangyin preparation were mostobvious(P<0.05),and there is no obvious difference significantly betweenthem(P>0.05).ConclusionsPreparation of Yiqi-huayu-huatan-yangyin preparation may protect theliver and delay the development of liver fibrosis by regulating the mRNAand protein expression of FSP1and PAR-2.
