Objective:To established rat models of abortion through the tail vein injection of lipopolysaccharide (LPS), and intervened with progesterone and LPS-RS, the embryos absorption rate is calculated and to detect IL-1β, TNF-α factor protein expression in decidua of abortion rats, Thus to explore the pathogenesis of septic abortion and the role of progesterone and LPS-RS in infection abortion.Methods:Male and female rats were fed on the same cage and the day when the vaginal plug was found in the female rats defined as the zero day of pregnancy. The pregnant rats were randomly divided into five groups to establish rat models:the first group was the control group which had the tail vein injection of phosphate buffer (PBS) lml each rat in the seventh day of pregnancy; the second group was the LPS group (abortion group) which had the tail vein injection of (LPS) lml each rat in the seventh day of pregnancy; the third group was the LPS+progesterone group which had the tail vein injection of LPS1ml each rat in the seventh day of pregnancy and intramuscular injection of progesterone0.2ml each rat in the sixth to eleventh days of pregnancy; the fourth group was the LPS+LPS-RS group which had the tail vein injection of LPS-RS lml each rat in the seventh day of pregnancy and followed by tail vein injection of LPS lml a hour later; The fifth group was the LPS+progesterone+LPS-RS group which had the tail vein injection of LPS-RS1ml each rat in the seventh day of pregnancy and followed by tail vein injection of LPS lml a hour later, and Additionally had the intramuscular injection of progesterone0.2ml each rat in the sixth to eleventh days of pregnancy. All rats were put to death on the thirteenth day of pregnancy and taken the decidua of the abortion rats to calculate embryo absorption rate. Comparing the differences of embryo absorption rate among groups and using western blot to detect the protein expression of IL-1β, TNF-α in the decidua of rat.Results:(1)The embryo absorption rate in the five groups were6.58%、24.79%、16.30%。9.89%、13.48%. The embryo absorption rate of the second group was higher than that of the first group and rat model of abortion was successfully established. The embryo absorption rate of the third, fourth and fifth group was higher than that of the first group but the differences were not statistically significant. The embryo absorption rate of the fourth and fifth group was lower than that of the second group and the differences were statistically significant. The differences among the third, fourth and fifth group were not statistically significant.(2)Using western blot to detect the protein expression of IL-1β and TNF-a in the decidua of rat:the protein expression of IL-1β and TNF-α in the second group was higher than that in the first and fifth groups and The differences were statistically significant (P<0.05). The protein expression differences of IL-1β and TNF-a among the third, fourth and fifth groups were not statistically significant(P>0.05).Conclusion:LPS successfully induced abortion in the pregnant rats and the pretreatment of LPS-RS and progesterone could lead to embryo absorption which caused by LPS. The protein expression of IL-1β, TNF-α were increased in decidua of. abortion rats, but using progesterone and LPS-RS makes these two cytokines down-regulated, suggesting that hormone therapy combined anti-inflammatory have better effect of septic abortion.
