Objectives:To testify the relationship on islet beta cell apoptosis by signal molecules of PDX-1, PPARs, PDX-1.Methods:MIN6cells had been treated with0.5mM AICAR (AMPK agonist) and10μM BML-275(AMPK antagonist) for24hours, nothing treated cells were set as control group, the mRNAs expression of AMPK, PPARa, PPARy, PDX-1were detected by Real-time quantitative PCR. Then MIN6cells were treated as following:Con, control group; group M,5μM MK886(PPARa antagonist); group B,50μM BADGE(PPARy antagonist); group AM,0.5mM AICAR+5μM MK886; group AB,0.5mMAICAR+50μM BADGE. Using Real-time quantitative PCR measured PDX-1mRNA level, the protein expression of P53, Caspase3was by Western blot method.Results:1. After AMPK agonist treated, AMPK, PPARa, PPARy, PDX-1mRNA were markly increased compared to control MIN6cells; while BML-275induced AMPK inactivation, AMPK, PPARa, PPARy, PDX-1mRNA expression were obviously reduced;2. Treated with PPARa/PPARyantagonist, PDX-1mRNA content was markly decreased in group M, B, AM, AB compared to control group, and failed to exam significant difference between group M and AM, between group B and group AB.3. The protein expression about P53and Caspase3was obviously elevated in group M, B, AM and AB compare to control group, but no difference between group M and AM, between group B and group AB.Conclusion:1. AMPK regulated PDX-1expression through PPARs.2. AMPK-PPAR-PDX-1and islet beta cells apoptosis are related.
