Objective:The purpose of this experiment is to test the ROCK-2and MMP-9expression in colon cancer tissues and cancer adjacent tissues, and explore these two indicators and the relationship between colon cancer clinical pathological features as well as the potential relationship between this two indicators, so as to provide some molecule theory basis for the treatment of colon cancer.Methods:78colon cancer tissues and para-carcinoma tissue which have been resected via operation from Jun to Dec,2013, were selected. The expression of ROCK-2and MMP-9were measured by immunohistochemical method, and next analyzed the difference of expression of ROCK-2and MMP-9in colon cancer tissues and para-carcinoma tissues. Then investigated relations between ROCK-2and MMP-9in the clinical and pathological data, including age, sex, tumor location, cell differentiation, CEA, serosal invasion, TNM stage, lymph node metastasis and distant metastasis. All statistics were performed using GraphPad Prism5.0software, completed with statistical significance(P<0.05).Results:1.Compared to para-carcinoma tissue, the expression of ROCK-2was higher in colon cancer tissues (P<0.05). The positive rate is64.1%and7.7%respectively; and compared to para-carcinoma tissue, the expression of MMP-9was higher in colon cancer tissues (P<0.05). The positive rate is71.8%and12.8%respectively. 2. The expression of ROCK-2in tumor tissues from which the patients with serasal invasion, Ⅲ+Ⅳ stage, lymph node metastasis and distant metastasis, was higher than the opposite tissues (P<0.05). The expression was correlated with invasive depth, lymph node metastasis, distant metastasis; and the expression of MMP-9in tumor tissues from which the patients with serasal invasion,Ⅲ+Ⅳ stage, preoperative elevated CEA group, lymph node metastasis and distant metastasis, is higher than that in the opposite tissue (P<0.05). The expression was correlated with CEA, invasive depth, lymph node metastasis and distant metastasis.3. The expression of ROCK-2and MMP-9in tumor tissues were not correlated with age, sex, differentiation and tumor location.4. There is a positive correlation between the protein expressive level of MMP-9and ROCK-2(R=0.364, P<0.05).Conclusion:1.The expression of ROCK-2and MMP-9are up-regulated in colon cancer tissues, promoting the development of Colon Cancer.2. ROCK-2and MMP-9were involved in invasion and metastasis of colon cancer, which promising to be potential targets for Colon cancer therapy.3.There is a positive correlation between the expression extent of MMP-9and ROCK-2and may promote the invasion and metastasis of colon cancer by a certain signaling pathway.
