Objective:This study was aimed at investigating the association of CYBA gene polymorphisms and cerebral hemorrhage in Changsha area. And further analyze the interactions between polymorphisms and risk factors.Method:this is a case-control study, a total of760cases and748controls were enrolled from January2008to December2013in xiangya hospital. The overall cerebral hemorrhage(CH) group was divided into four subgroups:①deep cerebral hemorrhage(DCH)535subjects;②lobar cerebral hemorrhage(LCH)125subjects;③infratentorial hemorrhage71subjects;④ventricular hemorrhage29subjects.DNA was extracted from blood using the traditional phenol-chloroform. TaqMan probe was used for genotyping and SPSS18.0for statistics analysis. We also used logistic regression and Andersson’s model of biological interaction to further analyze the interactions between polymorphisms and risk factors.Results:1.The frequencies of T allele of CYBA gene rs4673was0.067in CH group, and0.066in DCH, which were both significantly lower than that in the control group(0.088,P=0.035and0.049respectively). 2. No significant difference was found in the distribution of G allele of CYBA gene rs9932581polymorphism between CH group and controls(0.369and0.362respectively, P=0.671).The frequency in DCH was0.421,significantly higher than that in controls(P=0.002).3.The analysis of the effect of CYBA gene rs4673,rs9932581polymorphisms on blood pressure in changsha han Chinese found that: The systolic pressure of rs4673polymorphism CT/TT genotype carriers was significantly lower than that of CC genotype(P=0.012),while no difference was found between diastolic pressure(P=0.083).And no difference was found in systolic and diastolic pressure among different genotypes of rs9932581polymorphism(P=0.386and0.954respectively).4. The analysis of the effect of CYBA gene rs4673,rs9932581polymorphisms on the location of hematoma found that:the distribution of rs4673polymorphism was similar among all four subgroups(P=0.909).but the distribution of rs9932581G allele differed among DCH group,LCH group, infratentorial hemorrhage group and ventricular hemorrhage, the frequency was0.421,0.224,0.261,0.293respectively, P<0.01.5. The multiple logistic regression model of DCH was established, rs9932581G allele, systolic pressure, diastolic pressure, smoking history, BS,TG,HDL entered the final equation, the OR was1.651、4.403、1.850、3.213、4.719、1.971、5.210respectively. Interaction analysis found that there was positive interaction between G allele and low HDL, no interactions were found between G allele and other risk factors.Conclusions:1. CYBA gene rs4673polymorphism T allele might be a protective factor for cerebral hemorrhage, the carriers share a lower level of systolic pressure may be one of its possible mechanisms.2. CYBA gene rs9932581polymorphism G allele might be a risk factor for deep cerebral hemorrhage. There is an interaction between rs9932581polymorphism G allele and low-HDL, and the interaction might influence individual predisposition to deep cerebral hemorrhage.
