Objective: This study was aim to investigating the association of TNF-? gene variation and cerebral hemorrhage in Changsha han populati-on.Method: In this case-control study,a total of 162 cases and 171 controls were enrolled from January 2016 to September 2017 in Xiangya hospital.Patient with cerebral hemorrhage were diagnosed based on CT or MRI.Controls were recruited from health check-up during the same period.The overall cerebral hemorrhage(CH)cases group was divided into two subgroups:1.?deep cerebral hemorrhage(DCH):124 subjects,the bleeding structure including the basal ganglia,brain periventricular white matter,internal capsule,thalamus,brain stem,cerebellum.?Lobar hemorrhage(LH):38 subjects.including cortex and the anderlying white matter of cerebral hemisphere.2.The cerebral hemorrhage and the control groups were divided into TT and TC + CC subgroups,GG and GA + AA subgroups according to-1031 T/C Polymorphic genotypes and-308 G/A polymorphism.Ligase Detection Reaction(LDR)was used for genotyping and SPSS 19.0 for statistics analysis.We also used logistic regression model to further analyze the risk factors of CH.Results: 1.No significant difference was found in the distribution of T-1031 C genotype(TT and TC+CC)of TNF-?gene promoter polymorphism between CH group and controls(P=0.320),The frequencies of C allele also didn't show distinct difference between the two group(p=0.426).2.No significant difference was found in the distribution of G-308A genotype(GG and GA+AA)of TNF-?gene promoter polymorphism between CH group and controls(P=0.424),The frequencies of A allele also didn't show distinct difference between the two group(p=0.540).3.Analyzing the genotype and allele distribution of T-1031 C and G-308 A of TNF-?gene promoter polymorphism,no significant difference was found between the group of DCH and LH(the value of P were 0.283,0.229 respectively).4.The analysis of the effect of TNF-?gene promoter T-1031 C and G-308 A polymorphism on blood lipid level,blood pressure and fasting plasma glucose in changsha han population.?Both in the CH group and the control group,there was no significant difference in the level of TG,TC and LDL-C among different subgroups of gene type of T-1031 C polymorphism(the value of P were 0.237? 0.754? 0.730).the level of TG of the GA/AA gene type of G-308 A was significantly higher than that of the GG gene type(the value of P was 0.020).there was no significant difference in the level of TC and LDL-C among different subgroups of gene type(the value of P were 0.769? 0.373respectively).?Both in the CH group and the control group,the level of systolic pressure and diastolic pressure of the TT gene type of T-1031 C was significantly higher than that of the TC+CC gene type(the value of P were 0.030?0.045respectively);the level of systolic pressure and diastolic pressure of the GA+AA gene type of G-308 A was significantly higher than that of the GG gene type(he value of P were 0.023? 0.010 respectively).?There are no significant difference of fasting plasma glucose level was found between different genotype of T-1031 C and G-308 A polymorphism(p>0.05).5.The multiple logistic regression model of CH was established,the gender,alcohol intake,hypertention,high systolic pressure,fasting plasma glucose(FPG and high triglyceride level entered the final equation,the OR was 0.261?0.383?3.037?2.859?3.517?4.002 respectively.6.The multiple logistic regression model of DCH was established,the gender,smoking history,hypertention,high systolic pressure,high diastolic pressure,fasting plasma glucose(FPG)and high triglyceride level entered the final equation,the OR was 0.209?0.409?2.495?3.091?1.772?3.127?4.356 respectively.Conclusions: 1.There is no significant association of T-1031 C and G-308 A polymorphism of TNF-?gene promoter with cerebral hemorrhage and DCH in changsha han population.2.The level of systolic and diastolic pressure may be lowered by T-1031 C polymorphism,and the TG,systolic and diastolic pressure may be elevated by G-308A mutation.
