The Effect Of Fei Xin Decoction Drug Serum On Proliferation Of Rat Pulmonary Artery Smooth Muscle Cells And PHD2/HIF-1α/VEGF

【OBJECTIVE】Through Fei Xin Decoction(FXD) drug serum and PHD2siRNA combined treatmentPASMCs cultured in vitro, the experiment aims to explore the effects of FXD drug serum onrat PASMCs in vitro which proliferates in low oxygen conditions, PHD2, HIF-1α and VEGF.【METHODS】Firstly, lentiviruses transfect rat PASMCs cultured in vitro. Secondly, rat drug serum andnormal blank serum which were proved to contain effective Chinese medicine group of FXDculture rat pulmonary artery smooth muscle cells (PASMCs). Thirdly, the expressions ofHIF-1α、PHD2、VEGF、cyclinD1RNA are determined by Real-time PCR; protein expressions ofHIF-1α、PHD2、VEGF、cyclinD1are determined by Western-blot; different groups of cell cyclechanges are determined by ELISA.【RESULTS】1. The results of Real-time PCR and Western-blot show that LV-1can significantlyreduce PHD2MRNA and protein expression in the three groups of interferencesequence of PHD2.2. In low oxygen conditions, the results of Real-time PCR and Western-blot showthat comparing FXD drug serum treatment group with blank group, PHD2had nosignificant change(P>0.05), HIF-1α and VEGF expression were significantreduced(P<0.05).3. The results of Real-time PCR and Western-blot show that comparing FXT drugserum treatment group with blank group, cyclinD1expression reducedsignificantly; comparing FXT drug serum treatment group in cell cycle detectionwith blank group, proliferation index(PI) was markedly increased(P<0.05).【CONCLUSION】 1. FXD drug serum is involved in the regulation of PASMCs proliferation by PHD2regulating HIF-1α.2. FXD drug serum can reduce PASMC proliferation，reduce expression of HIF-1α/VEGF.
