This dissertation is composed of four chapters. The first chapter discussed the fat-soluble components in P.maximowiczii. by GC-MS. The second chapter included the antioxidant capacity of Phlomis maximowiczii Regel. Andthe hepatoprotective effect of P.maximowiczii. on acute liver injury induced by carbon tetrachloride in mice. The third chapter discussed the effect on blood coagulation of P.maximowiczii. and Psoralea coryylifolia L.. The fourth chapter summarized the activity of anticoagulant and antithrombotic of flavonoid phytochemicals from the plant.Chapter Ⅰ Analysis of the fat-soluble components in Phlomis maximowiczii by GC-MSThe fat-soluble components of petroleum ether extract from P.maximowiczii. were analyzed by GC-MS.12compounds from petroleum ether were identified, accounting for80.45%of the total fat-soluble components. Alcohol (33.37%) and esters (25.02%) were dominant in the fat-soluble components, the main compounds were γ-sitosterol (17.16%), stigmasterol (3.49%) and campesterol (4.73%).Chapter Ⅱ Hepatoprotective effect of P.maximowiczii.1. Antioxidant activity of different extracts of P.maximowiczii. was evaluated by scavenging1, l-diphenyl-2-picrylhydrazyl (DPPH) radical and [2,2’-azino-bis(3-ethylbenzothiazoline)-6-sulphonic acid] diamonium salt (ABTS) radical in vitro. The study discovered that n-butanol (PMBU) extract, ABTS (IC50=18.96μg/mL), DPPH (IC50=25.15μg/mL) and FRAP (RACT50=2775.6±144.18μmol/g), showed higher scavenging capacity than that of P. maximowiczii. ethyl acetate (PMEA). In DPPH assay, the antioxidant activity of PMBU (IC50=25.15μg/mL) was lower than that of BHT (IC50=18.71μg/mL). In FRAP assay, the antioxidant activity of PMBU (RACT50=2775.6±144.18μmoL/g) was higher than that of PG (RACT50=1581.68±97.41μmoL/g). The results showed that the antioxidant activity of PMBU had the highest antioxidant activity in vitro.2. Hepatoprotective effect of P. maximowiczii. extracts was evaluated using carbon tetrachloride-induced acute liver injury in mice, the levels of GOT, GPT, SOD, and MDA were detected to evaluate protective effects of ethyl acetate and n-butanol extract from P.maximowiczii.. Results showed that a significant in the level of GOT and GPT in CCl4-treated mice (P<0.001). Levels of GPT and GOT, MDA decreased significantly (P<0.05and P<0.01and P<0.001, respectively) and each dose group of PMBU could increase the level of SOD (P<0.001) respectively. The result indicated that the antioxidant activity of n-butanol extract in vitro had relationship with the level of MDA and SOD in vivo, and hepatoprotective effect of n-butanol extract also had relationship with its antioxidant activity in vitro.Chapter Ⅲ Effects of the Phlomis maximowiczii Regel and Psoralea coryylifolia L. on Blood Coagulation1. The effect of extracts of P. maximowiczii and Psoralea coryylifolia L. and8compounds of P. coryylifolia on blood coagulation was investigated using Vitamin Ki as coagulant control and breviscapine as anti-coagulant control. n-Butanol extract of P.maximowiczii and P. coryylifolia and ethyl acetate extract of P.maximowiczii significantly prolonged the plasma recalcification time (P<0.001). Corylifolin, daidzin and daidzein significantly reduced the plasma recalcification time (P<0.001). Isopsoralen and psoralidin significantly prolonged the plasma recalcification time (P<0.001).2. By establishing the fibrinolytic pathway inhibitory model in vitro, the fibrinolytic inhibitory activities of different extracts from P.maximowiczii and P. coryylifolia and8compounds of P. coryylifolia were screened by fibrin plate method. Results showed that n-Butanol extract from P.maximowiczii had the highest inhibitor activity (IC50=1.5962mg·mL-1), and followed by ethyl acetate extract (IC50=3.0506mg·mL-1). Ethyl acetate extract from P. coryylifolia had the highest inhibitor activity (IC50=2.687mg·mL-1), and followed by n-Butanol extract and petroleum ether extract (IC50=2.7675and4.6051mg·mL-1). The activities were higher than that of Compound Danshen Tablets (IC50=6.35621mg·mL-1) as positive control. The sequence of the inhibitor rate of8compounds from P. coryylifolia is as follows:corylifolin (50.44)> psoralidine (49.83%)> daidzin (20.25%)> daidzein (16.36%)> isopsoralen (3.62%).8-Methoxypsoralan and psoralen showed a weak activity of promote fibrinolysis. Chapter Ⅳ Research progress of anticoagulant and antithrombotic of flavonoid phytochemicals from the plantThe anticoagulant and antithrombotic activity research progress of inhibition of tissue factory, inhibition of platelet aggregation, promoting the fibrinolytic system and protecting endothelial cells were summarized.
