The flavonoids content in aerial parts of above-ground licorice is at a high level and flavonoids can induce a certain degree anti-inflammatory and anti-tumor effects.To make full use of liquorice resources,we should step up pharmacological research of flavonoids in aerial parts of above-ground licorice.Previous studies have found that aerial part of above-ground licorice shows good anti-prostatitis activity,but its main efficacy material base is still unknown,to make full use of above-ground licorice,it’s extremely important to find its efficacy material base.Due to differences of solvent,wo got different kinds and content of flavonoids,our study adopted two kinds of solvents,water mention and alcohol extraction,after which the alcohol extract extraction solvent recovery to no alcohol taste,we take the supernatant.But other non flavonoids impurity in the aerial parts of above-ground licorice hinder the further use of aerial parts of above-ground licorice,therefore there is a need in aerial parts of above-ground licorice enrichment and purification of water-soluble flavonoids.Based on this,our study was mainly about different sections of the supernatant of taerial parts of above-ground licorice and screening of the active substances,as well as inspecting aqueous extract of total flavonoids enrichment path of the aerial parts of above-ground licorice;we also take a comprehensive evaluation of antioxidant activity of water-soluble flavonoids and evaluation of anti-prostatitis activity of water soluble flavonoids of aerial parts of licorice different based on different segmented parts,all these work lay a solid foundation for the future development of aerial parts of above-ground licorice.The results are as follows:1、The supernatant fluid of aerial parts of above-ground licorice will be preliminary qualitative divided into the area of 1 and 3Macroporous resin will be used to segment the supernatant fluid of aerial parts of above-ground licorice,and then we compared their characteristic spectrum with similarity,made combination of flavonoids content,yield and solid yield as the index for segmentation research.Due to different solvent elution ability,we first selected three gradient that can be segmented,then took previous experimental results and the experiment purpose into comprehensive consideration,chose feasible further screening way between the two methods,took flavonoids content yield,yield,and solid for comprehensive consideration,it will be feasible that the supernatant fluid of aerial parts of above-ground licorice will be divided into the area of land 3.We conducted preliminary qualitative results of the supernatant fluid of aerial parts of above-ground licorice through HPLC-MS to find out the control peakof the rutin,quercetin and rhizoma kaempferiae phenolic glucoside.2、Optimizing a set enrichment technology of water-soluble flavonoids of aerial parts of above-ground licoriceWe took preliminary verification of anti-inflammatory activity of the supernatant fluid of aerial parts of above-ground licorice,the experimental results showed that the supernatant fluid of aerial parts of above-ground licorice has better anti-inflammatory activity,it is worth further study.Taking adsorption and desorption rate of total flavonoids as index,we selected the best macroporous resin model,and inspected its sample-adsorption-elution parameters of the process,we sampled the supernatant fluid of aerial parts of above-ground licorice(pH=5.42)with a mass concentration of 2.128 mg.ML-1,volume for 5 BV,above 1.5 BV/h on sample.After 1 h,we successively eluted 2 BV water,4 BV 60%ethanol.Results showed that rhe total flavonoids raised 9.08%to 34.42%,a 3.8-fold increase purity,the yield was 74.40%.Th en we got the a conclu sion:using SZ6 resin to purificate flavonoids in aerial parts of above-ground licorice is relatively stable.3、The area of 3 showed better antioxidant activity and can be fully utilized in future.With ascorbic acid and rutin as positive control,we took four different principles of antioxidant test to compare the antioxidant activity at different concentrations of licorice extract of aerial parts of water,the supernatant water extract of flavonoids and 1 of sites and 3of aerial parts of above-ground licorice.3 parts showed the best antioxidant activity,while the parts of the supernatant fluid is the worst.Aqueous extract flavonoids showed stronger antioxidant activity than aqueous extract,and with the increase of flavonoids content,its antioxidant activity rises as well.Antioxidant activity increased with the increase of concentration of sample.Therefore,we should intensify enrichment and purification of total flavonoids of aerial parts of above-ground licorice.4、3 parts of the SAPL showed best antiprostatitis activity and can be fully utilized in future.In this study,we used chemical agents to induce inflammatory in order to create a model of Wistar rats of prostatitis,measured its organ and serum indexs,HE was taken to observe the histopathologic changes of rats in different groups and Masson was used to measure the degree of fibrous tissue hyperplasia,immunohistochemical studies in the course of disease to detect five important cytokines:TNF-α、MCP-1、TGF-β、CTGF、α-SMA.The experimental results showed that water soluble flavonoids of the aerial parts of licorice has good antiprostatitis activity and 3 parts is the best one.
