Some familiar traditional Chinese medicines were selected. The antioxidant activities were evaluated by using the Oxidative Stability Instrument (OSI) in lard at 100℃ to screen the natural antioxidants. The antioxidant activity is showed as Pf. Inthe crude herbs of 65 traditional Chinese medicines, 9 were found to have significant antioxidant effect on the lard. Their antioxidant activities decrease as follow order: Rosmarinus officinalis L., Psoralea corylifolia L., Myristica fragrans Houtt., Glycyrrhiza uralensis Fisch, Rhus chinensis, Terminalia chebula Retz, Salvia miltiorrhiza Bge, Syzyglum aromatioum merr (Pf>2). The antioxidant activities of Psoralea corylifolia L. were stronger than chemical antioxidants BHA, BHT and natural antioxidant a-tocopherol. It is a widespread herb. Few reports, however, have addressed the antioxidant activity of Psoralea corylifolia L. So it may be worth of studying as a potential antioxidant resource. On the basis of systematical consulting the researching literatures of chemical constituents and antioxidant activities study of Psoralea corylifolia L, and completing literature summary, by means of antioxidant-guiding, six compounds were isolated from the herb. They were identified by UV, IR, Mass,1H and 13C-NMR spectra and physical-chemical properties. They are bakuchiol, psoralen, isopsoralen, corylifolin, corylin and psoralidin. The antioxidant properties of these compounds were investigated individually and compared with those most commonly used antioxidants , BHT and a-tocopherol by using OSI in lard at 100℃. Their action mechanisms were also preliminarily explored. The results showed that bakuchiol, corylifolin, corylin and psoralidin had strong antioxidant activities, especially psoralidin had stronger antioxidant property than BHT at 0.02% and 0.04% levels, but psoralen and isopsoralen had no antioxidant activities at 0.02% and 0.04% levels. The antioxidant activities of the compounds decrease as follow order: psoralidin > BHT >a-tocopherol > bakuchiol > corylifolin > corylin > isopsoralen ~ psoralen. These findings suggest that the antioxidant activities of phenols closely relate to the environment conditions of phenolic hydroxyl groups rather than the numbers of phenolic hydroxyl groups. In the experimental research, the antioxidant activities and anti-aging of Psoralea corylifolia L. was performed in vivo for the first time. The content of the activities of SOD and MDA were measured in the blood serum of mice. With a-tocopherol as the positive control, the effect of Psoralea corylifolia L. was observed on the swimming time and the survival time under the conditions of normobaria hypoxia and hyperthermia in mice. The life of Drosophila melanogaster was measured afterwards. The results showed it could lessen fatigue, anti-hypoxia and anti-hyperthermia of mice and prolong the mean life, median lethal life and longest life of Drosophila melanogaste. It also evidenced that it could prolong life and improve the quality of life. The study examined the antitumour effects of the active compounds in vivo. The influence of ethanol and dimethyl sulfoxide on cell proliferation assay was also investigated. The study examined the growth inhibitory effects of the active compounds on BGC-823, U937, Du-145, A-549, Bel-7402 and Eca-109 human cancer cells by MTT. To evaluate the inhibitory effect of psoralen and isopsoralen on the proliferation of human cancer cell lines, exponentially growing BGC-823, U937, Du-145, Bel-7402 and Eca-109 cancer cells were cultured in 10% BCS medium containing different amounts of the compounds for 48 h. Psoralen and isopsoralen exhibited a differential growth-inhibitory effect. IC50 values of psoralen and isopsoralen respectively were 5.820 ug/mL, 32.64 ug/mL, 29.83 ug/mL, 48.81 ug/mL, 23.85 ug/mL, >100 ug/mL and>100 ug/mL, 59.53 ug/mL, 69.19 ug/mL, >100 ug/mL, 39.74 ug/mL, >100 ug/mL. The effects of psoralen and isopsoralen on different cancer cells were increased as the following order: BGC-823 > Bel-7402 > Du-145 > U937 > A-5...
