Veratrum dahuricum (Liliaceae) is a perennial herb. Chinese medicine Veratrum includes a variety of plants of the genus hellebore, distribution around the world is about more than40, widely distributed in the northern hemisphere temperate regions. Its taste hard, there are wind Chung spit phlegm, pharyngitis nowhere, detoxification and insecticidal effect. Chinese medicine for the treatment of stroke aphasia and phlegm, jaundice, chronic malaria, pharyngitis, breath, drainage flow field, headache, scabies, malignant sore and so on.In recent years, studies have shown that Veratrum dahuricum mainly contains Verazine type, Jervine type, Solanidine type and Cevanine type. It has the pharmacological effects such as lowering blood pressure, antithrombosis, strong heart, improving brain circulation, resistancing to parasitic infections and so on. At the same time, it have obvious toxicity on the respiratory system, cardiovascular system and nervous system.At present, although studies on the constituents and pharmacological activities of Veratrum dahuricum, it restricted to market development and clinical application for its toxicity. And its toxicology mechanism is not clear, so it is still insufficient to the depth and breadth research. We hold the opinion that no matter from the study of chemical composition or pharmacological activity research perspective, it must have more thorough system research on the active ingredient. Hellebore alkaloids are both active ingredients and toxic ingredients, only for preparation of large numbers of hellebore alkaloids to provide sufficient data for their toxicity research and material, regarding the smooth conduct of experiments. Veratrum dahuricum as a kind of the Veratrum Linn., it has different compositions from other kinds of Veratrum. This experiment used many kinds of separation and purification methods to obtain its chemical compositions, and combined with the spectral data identified compound structure for the next step of pharmacology and toxicology experiments to provide experimental materials and data.Since previous studies found that alkaloids can cause DNA damage in mouse cerebellum and cerebral cortex, and the extent of DNA damage is also different for the different structure. This paper studied the chemical composition of the Veratrum dahuricum. By silica gel column chromatography, dextran (Sephadex LH-20) chromatography, HPD100macroporous resin and semi-preparative chromatography, high performance liquid chromatography method for the Veratrum isolated compounds were extracted to obtain single donor compound. In combination with1H-NMR,13C-NMR, HMBC, HMQC, ESI-MS spectra analysis was carried out on the compound and the physical and chemical reaction in the method of identification. The experiment from Veratrum album ethanol extract by extraction and separation and purification systems, were isolated and identified10kinds of monomer compounds.They are verussurinine (1), veratramine (2), veratroylzyade (3), veramitaline (4), mannitol (5), jervine (6), germerine (7)、7,4’-dimethoxy-4-hydroxy-auraone-6-0-β-D-glucosidase (8*)、2β-hydroxylverdine (9*), tomatillidine-3-O-β-D-glucopyranoside (10*). Which verussurinine (1) for the first time been isolated from Veratrum dahuricum. Mannitol (5) is first isolated from Veratrum plants.7,4’-dimethoxy-4-hydroxy-auraone-6-O-β-D-glucosidase(8*).2βhydroxylverdine(9*) and iomatillidine-3-O-β-D-glucopyranoside(10*) are new compounds.
