Effects Of The Regenerating Liver Phosphatase3on The Metastasis And Proliferation Of Glioma Cell

There are many literatures report that Phosphatase of regenerating liver3(PRL3)promotes proliferation, invasion and metastasis of variety tumor cell, but the detailmechanism and effect on glioma cells remain unclear. In this research we focus on therelationship between PRL3and glioma cells.Objective: Identify the proteins binding to PRL3protein and provide evidenec forthe further study of the molecular mechanism of PRL3in the signal pathways. Research theeffect of the overexpression and silence of PRL3on the proliferation and migration ability ofgliomas cells.Methods: Form prokaryotic express vector pGEX-6P-1-PRL3and induce theexpression of the fusion protein with GST tag, screen for the protein interaction with PRL3incells by pulling-down and purification and identify the novel proteins by mass spectrum.Form eukaryotic express vector pcDNA3.1-PRL3and transfected them into glioma cell withthe comparison of pcDNA3.1; Synthetic siRNA and form recombinant plasmidsiRNA-pSilencer3.1to make gene silencing system in U251cells with the comparison ofpSilencer3.1. The ability of the proliferation and migration of cells were measured.Results:(1) The result of pull-down shows four novel protein bands and identifiedone of them is tubulin.（2）Compared with the control, the number of cells with PRL3overexpressed were increased by cell counting; the number of the PRL3silenced glioma cellsdecreased but there was no significant changes in migration compared to control cells.Conclusion: We filtration for proteins in the brain of mouse and one of them hasbeen identified to be tubulin-beta. The overexpression of PRL3promote proliferation andmigration of glioma cells and the silence of PRL3inhibit the proliferation of glioma cells, but there is no significant effect on the cell migration.
