Objective To detect the differences in the expression of DOK7 gene in human glioma tissues and normal brain tissues,and apply the plasmid of the DOK7 gene recombination to transfected human glioma U87 cells,increase the m RNA and protein expression,to explore its effect on human glioma cell proliferation.Methods Using polymerase chain reaction(PCR)technology to detect DOK7 in human glioma,glioma cell line U87 and normal brain tissue(get)by brain trauma surgery in the expression of m RNA level,and by using Western blot test(Western blot method)to detect the expression of its coding protein;Through DOK7 gene recombinant plasmid vector transfection human glioma U87 cells,resulting in increased expression DOK7 m RNA and protein of glioma cells(U87),color test using tetrazolium salt(determined by MTT colorimetry)detect the proliferation ability of change;The effect of the cloning of DOK7 plasmid on the cloning of glioma cells was investigated.Results PCR technology detection tip: the mRNA expressed by DOK7 gene in glioma tissue was significantly lower than that of m RNA expressed in normal brain tissue(P < 0.01).Western blot test showed that the protein expressed by DOK7 gene was significantly reduced in glioma tissues(P < 0.01).After the DOK7 plasmid transfection of U87 cell line of human glioma,the expression of m RNA and encoded protein in the transcriptional transcription was significantly increased(P < 0.01).Determined by MTT method to detect shows: after DOK7 plasmid transfection of glioma cell proliferation significantly decreased(P < 0.01),flat cell clone formation experiment also prompt DOK7 plasmid of glioma cells after transfection,the ability to clone formation decreased obviously(P < 0.01).Conclusion DOK7 genes are expressed in human glioma tissue in the normal brain tissue decreased obviously,by plasmid transfection technology increase DOK7 gene expressed in the glioma cells can significantly inhibited the proliferation of glioma cells and cloning formation ability,after the discovery of the molecular biology level provides a new path for the inspection and diagnosis and treatment of glioma.
