Objective: To investigate the influence of Total flavone of LitchiChinensis Sonn (TFL) on expression of TGF-β1and OPN in fibrotic liverand its therapeutic effect on Cholestatic liver fibrosis in rats.Methods:Total100males SD rats were randomly divided into five groups:sham-operated group, model group, high-dose TFL group (300mg·kg-1·d-1),mid-dose TFL group, low-dose TFL group.The model with liver fibrosis wasinduced by common bile duct ligation in rats, whereas the sham-operatedgroup was used as controls. The rats were sacrificed at the end of2nd week and4th week after operation. Serum level of hyaluronic acid(HA), laminin (LN), procolagen type Ⅲ(pcⅢ) and Ⅳ collagen (Ⅳ-c)was detected by radioimmunoassay, HE and Masson staining wereconducted in hepatic tissues to observe pathological variations as well, Theexpression of TGF-β1and OPN was determined by immunohistochemistry.Results: Serum Level of HA, LN, pcⅢ and cⅣ of sham-operated group, high-dose TFL group and mid-dose TFL group was siginificantly lower thanthose in model group and low-dose TFL group at the end of2rd week and4thweek after operation (P<0.05), so it was with the expression levels ofTGF-β1and OPN in liver tissue. However, there was no obvious differenceamong the sham-operated group, high-dose TFL group and mid-dose TFLgroup at the end of2rd week and4th week after operation(P>0.05), therewas also no significant difference between model group and low-dose TFLgroup at the same time(P>0.05). Conclusion: High-dose TFL andmid-dose TFL may effectively reduce the liver injury and hepatic fibrosisinduced by common bile duct ligation. Its mechanism may correlated withinhibiting over-expression of TGF-β1and OPN and reducing the secretion ofECM.
