Obstract: Observed the expression levels of the TGF-β1/Smadsignal path key signaling molecule transforming growth factor-β1(TGF-β1), Smad3, Smad7in Spargue-Dawly (SD) rats’ livertissues,which were dimethylnitrosamine-induced liver fibrosis andintervened by TFL (total flave one from Litchi chinensis Sonn).Andexplore the antifibrotic mechanism of TFL.Methods: The liver fibrosismodel was induced by intraperitoneal injection of dimethylnitrosamine(DMN) in SD rats for4weeks and were intervened by the way of TFLintraperitoneal injection (different dose:400,200,100mg kg-1d-1) for6weeks.And Compared with the Silybin group as the positive controlgroup. On the sixth week, to detected aspartate transaminase (AST),alanine aminotransferase (ALT),venous were siphoned from inferiorvena.To detected the pathological changes and liver fibrosis, Specimens from the same liver parts were observed by hematoxylin-eosin (HE)staining, Masson staining and the expression of TGF-β1, Smad3, Smad7mRNA were detected by RT-PCR(Reverse Transcription-PolymeraseChain Reaction).Result: Compared with blank control group, as thedegree of liver fibrosis in rats increased in model group, and the ASTand ALT in the serum significantiy increased, the expression of TGFbeta1and Smad3mRNA in the liver tissue obviously increased (P <0.05), while the expression of Smad7mRNA significantly reduced (P <0.05),Compared with model group, the serum AST, ALT in each dosegroup with joining TFL and silibinin group were significantly lowerthan the model group, the expression of liver TGF-beta1, smad3significantly decreased (P <0.05), while the smad7was increased.Conclusion: TFL may reduce liver injury and improve liver fibrosis inSD rats, and it may be closely related with suppression the expression ofTGF-β1, Smad3mRNA, and increased the expression of Smad7mRNA.
