Objective: Determination of brain glioma in vitro sensitivity tochemotherapeutic dargs by MTT method, which provides reference for thedesign of clinical individual chemotherapy.Methods: Based on22cases of postoperative pathological examination and inexpeirments using immunohistochemical methods were confirmed as freshglioma specimens,Preparation of single-cell suspensions of tumor pirmaryculture in vitro, and clinical cancer chemotherapy drug commonly usedconcentrations (by peak plasma concentration (PPC)) for72hours. MTT methodwas used to detect the different individuals of different chemotherapy drugs ontumor and their mutual two pairs of the sensitive and resistant.Results: The sensitivity of22cases of brain glioma samples to5chemotherapeutic drugs paired off in the order of THP+Vm-26> BCNU+Vm-26> ADM+Vm-26> Vm-26+DDP> THP+BCNU> ADM+BCNU>BCNU+DDP> THP+DDP〉ADM+DDP> THP+ADM, and to5chemotherapeutic drugs alone from high to low are Vm-26〉BCNU>THP〉ADM> DDP. Age, gender has no effect on the majority of drug sensitivity,tumor grade only has an effect on the part of the drug sensitive rate. Conclusion: MTT assay in vitro susceptibility testing is a simple, economical,accurate susceptibility testing,it plays an important role in clinical reasonablydevelop individualized chemotherapy? Five cases were resistant to commonlyused anticancer drugs, sensitivity vaires, in medication single, the Vm-26is best,and CDDP is one of the most powerful drug-resist of tumor cells.but atfer thechemotherapy drugs pairwise matching combination can significantly improvethe tumoricidal effect, which Vm-26+THP combination is the most obviouseffects,but THP+ADM combined experiment does not improve drugsensitivity, and no significant difference in mono-therapy.
