Objective According to the test about vitro chemotherapeutic sensitivity of primary culture cells of spongiocytoma and the expression of MGMT, MRP and P-gp, to explore the correlation between the sensitivity of chemotherapeutics to the brain glioma cells and patients'age, sex and the expression of drug resistance protein in gliomas, so as to establish methods that can predict the chemotherapeutic sensitivity in gliomas and supply theoretical instructions for designing optimal individualization chemotherapy to glioma patients.Methods Eighty fresh specimens of spongiocytoma were obtained and then carried out vitro primary culture. The MTT assay was used to determine the chemotherapeutic sensitivity of vincristine(VCR), cisplatin(DDP), nimuline(ACNU), temozolomide(TMZ) and teniposide(VM-26); the method of immunohistochemistry was used to determine the MGMT, MRP and P-gp expression in 64 clinical spongiocytoma specimens and 10 normal brain samples.Results The sensitive rates of VCR,DDP,ACNU,TMZ and VM-26 in 64 clinical spongiocytoma specimens which were succeed in primary culture were 9.4%, 43.8%, 57.8%, 51.6% and 62.5%; VM-26 ranked the first, with statistical significance in the difference (P<0.05); the positive rates of MGMT,MRP and P-gp expression were 67.2%,78.1%和42.2%. It was significantly higher than that in normal brain samples; the positive expressions of MGMT,MRP and P-gp were not correlated with patients'age and sexuality significantly; the positive expressions among MGMT,MRP and P-gp were not correlated significantly too. The sensitive rate of the malignant spongiocytoma specimens expressed by MGMT positive to ACNU and TMZ is obviously lower than the negative; the sensitive rate of the specimens expressed by MRP positive to VCR,ACNU and TMZ is evidently lower than the negative; the sensitive rate of the specimens expressed by P-gp positive to ACNU and VM-26 is markedly lower than the negative.Conclusions The sensitivity of the same glioma to different chemotherapeutic agents and the same chemotherapeutic agent to different gliomas is different. Chemotherapeutic sensitivity determination used MTT assay can provide guidance to designing optimal individualization chemotherapy to glioma patients and avoid the blind application of resistant drugs. MGMT, MRP and P-gp are highly expressed in the malignant spongiocytoma. The positive expressions of MGMT,MRP and P-gp can improve the drug resistance of spongiocytoma to a large extent , and the study of them is helpful to improve the results of chemotherapy in gliomas. |
