ObjectivesTo study the quantitative assay of the preferentially expressed antigen ofmelanoma (PRAME) gene expression in myelodyoplastic syndromes by real timeRT-PCR and Explore the significance of PRAME gene in the diagnosis and prognosisof MDS.MethodsThe quantitative assay of PRAME gene expression by real time RT-PCR wasconducted in38initial diagnosed myelodyoplastic syndromes of the WHOclassification patients and11nonmalignant hematological diseases.Using SPSS17.0software for statistical analysis,P<0.05for the difference was statistically significant.Results1. The PRAME gene was expressed in21.1%of all38patients withmyelodyoplastic syndromes,the median is0.5456(0-0.9832), but was not expressedin the control group. 2. The PRAME expression of myelodyoplastic syndromes was significantlyhigher than the control groups in38MDS patients,there were8patients withPRAME expression (21.1%),the positive ratio of the PRAME patients inRCUD/RCMD/RAEB-1/RAEB-2were respectively22.2%,14.3%,16.7%.37.5%,whileother group were not expressed,there were no significant difference between groups(P=0.162),but the PRAME expression of RAEB was higher than other groups.3. There was no obvious difference of PRAME expression and positive rate inthe IPSS group(P=0.321).However, the patients in high-risk were higher frequencyof PRAME expression than those in low-risk groups(13.3%vs.33.3%,P<0.05).4. The PRAME expression was not correlated with age, gender, WBC,HGB,PLT(P>0.05).5.2patients with PRAME expression were successfully followed, the conditionand expression level of PRAME had no significant in one case with RA.After3months,the other with RAEB-2converted to acute myeloid leukemia-M2, and thegene expression level was Significantly higher than before.Conclusion1. The PRAME expression levels of MDS patients is significant higher thannon-hematopoietic malignancy patient,and the expression level varies with thedisease.2. There was no significant difference of PRAME expression and positive rate inthe MDS subgroup of WHO classification and IPSS group,However, the patients inhigh-risk were higher frequency of PRAME expression than those in low-risk groups.
