Objective: Early spontaneous abortion(ESA) is a common problem ofThe reproductive medicine,is one of Obstetrics and gynaecology commonpathological pregnancy, The cause involves genetics, endocrinology andimmunology, and many other factors. At the blastocyst success afterimplantation, cells and cells, cells with extracellular matrix adhesion and theinteraction at the blastocyst play an important role in the developmentprocess.Galectin-1and galectin-3is two important members of galectins,thatdistributed in a variety of organizations, participate in cell proliferation,differentiation and local immune adjustment. The function of galectin-1andgalectin-3in the reproductive field gradually be attention. Studies have foundthat galectin-1,3play an important role at the blastocyst implantationprocess. But in embryonic development and the pregnancy in the process ofmaintaining the effect is less. The purpose of this investigation was to detectthe expression of galectin-1mRNA, galectin-3mRNA and Related proteins inthe Normal endometrium,the decidua tissues of spontaneous abortion andnormal human abortion and to study the effect of they in nourish cellsinfiltration and the formation of the placenta. So we can provide certaintheoretical basis in early pregnancy prevention and treatment of abortion andassisted reproductive technology application and the follow-up studies.Methods: This study included30cases decidua tissues of spontaneousabortion and30cases decidua tissues of normal human abortion in thedepartment of Gynecology in the Second Hospital of Hebei MedicalUniversity from March in2010to March in2011.30cases of normalendometrium were obtained from patients who examined by hysteroscopy. Allthe patients had diagnosed by pathology that they are normalendometrium.Normal endometrium included20cases of Hyperplasia period and10cases of secretory.No patients had history of Taking hormone drugs,complications and Adverse pregnancy history.All specimen collectioncompleted in20minutes. All of these tissues were rinsed sufficiently byDEPC water and immediately frozen in liquid nitrogen. These tissues werestored-80℃until further detection. At the same time, a part of tissues werestored in4%of Formaldehyde solution for further detection. Reversetranscription polymerase chain reaction(RT-PCR)and Immuno-histochemicaltechnique were used to detect the expression of galectin-1mRNA,galectin-3mRNA and Related proteins in the Normal endometrium,thedecidua tissues of spontaneous abortion and normal humanabortion.Statistical analysis of the data was performed using the StatisticalProduct and Service Solution13.0(SPSS13.0).All the measured value wereexpressed as mean±standard deviation (X±S).The data of each factor meetnormal distribution.The t-test and ANOVA was adopted to analyze the meansof the expression of galectin-1mRNA, galectin-3mRNA and Related proteins.The correlation coefficient between galectin-1mRNA, galectin-3mRNA wasdetermined by linear correlation analysis.A value of P<0.05was consideredstatistically significant.Results:1The expressions of galectin-1,galectin-3were detected in the normalendometrium,the decidua tissues of spontaneous abortion and normalhuman abortion, But they were significantly higher in secretory phase ofnormal endometrium than that in proliferrative phase (P<0.05).2The expressions of galectin-1mRNA and protein were significantlyhigher in the decidua tissues of normal human abortion than thatboth in the decidua tissue of spontaneous abortion and in the normalendometrium. The expressions of galectin-1mRNA and protein weresignificantly higher in the decidua tissues of spontaneous abortion thanthat in the normal endometrium (P<0.05).3The expressions of galectin-3mRNA and protein were significantlyhigher in the decidua tissues of normal human abortion than that both in the decidua tissue of spontaneous abortion and in the normalendometrium. The expressions of galectin-3mRNA and protein weresignificantly higher in the decidua tissues of early spontaneous abortionthan that in the normal endometrium (P<0.05).4In the decidua tissues of normal early pregnancy The expressions ofgalectin-1mRNA and galectin-3mRNA were positivelycorrelated(r=0.565,p<0.05).Conclusion:1The expressions of galectin-1mRNA, galectin-3mRNA and relatedproteins were significantly higher in the decidua tissues of normalhuman abortion than that both in the decidua tissue of spontaneousabortion and in the normal endometrium. It illuminated that the galectin-1and galectin-3play a part in the development of the early embryonic andin the process of embryos implanting.and placenta forming.2The expressions of galectin-1mRNA, galectin-3mRNA and relatedproteins were significantly higher in secretory phase of normalendometrium than that in proliferrative phase. It illuminated that thegalectin-1and galectin-3may participate in the process of blastocystimplantation.3It is useful to forecast the development of spontaneous abortion bycombining to detect the expressions of galectin-1and galectin-3. So wecan provide certain theoretical basis in early pregnancy prevention andtreatment of abortion and assisted reproductive technology applicationand the follow-up studies.
