BackgroundThyroid gland is the largest endocrine gland in the human body,thyroid hormonesplays an important role in body’s growth and development,and participate in themetabolic activity of human tissues and organs.The dysfunction of thyroid seriouslyaffect the metabolism of body,cause the destroy of nervous system and cardiovascularsystem.the shape of thyroid,which is located in the front of trachea,compress ordamage the surrounding tissue,causing a series of clinical symptoms,when it differentfrom the normal.Thyroid cancer is the common malignant tumor in head and neck. Inrecent year,as one of the fastest rising malignant of women, the incidence of thyroidcancer showed a trend by the rate of14.51%,and more women thanmen,man/female=1/3.38;Thus there is no specific tumor molecular makers to behelpful for the diagnosis of thyroid cancer. IMP3(insulin-like growth Ⅱ mRNA-binding protein3, IMP3) is a number of insulin-like growth factorⅡmRNA bindingprotein family, the existing research shows that it is high expression in a variety ofcancers, and is closely related with the progress of the tumor, metastasis andprognosis. CD44v6is expressed in a variety of cancers and high, with the invasion oftumor growth, development and metastasis are closely related. At home and abroadfor IMP3expression in thyroid papillary carcinoma research less, confirmed by immunohistochemical method to detect the IMP3and CD44v6expression in thyroidpathological tissue level, which discuss IMP3and CD44v6expression in thyroidpapillary carcinoma and its clinical significance.ObjectiveStudy IMP3and CD44v6gene expression in benign thyroid tissue, thyroidpapillary carcinoma and papillary thyroid carcinoma with lymph node metastasis,and analysis the relationship between the two genes expression of thyroid cancer withclinical cases data, to explore the role of the two genes in the tumorigenesis and thedevelopment process of thyroid cancer,explore the correlation of the gene expression,discussed whether the both can be used as a specific molecular markers, and assist inthe diagnosis of thyroid papillary carcinoma.MethodSpecimens from January2013to August2013, the first affiliated hospital of zhengzhou university line of surgical treatment of70patients with thyroid, pathologicaltypes were confirmed by pathology, all specimens by4%formaldehyde fixed,paraffin embedding. Papillary carcinoma of thyroid follicular type (follicur variantsof PTC, FVPTC)10cases of non follicular papillary carcinoma,20cases; Thyroidpapillary carcinoma and20patients with lymph node metastasis;20patients withbenign thyroid tissue, including10cases of normal thyroid tissue (cut edge and nodaldistance of2.5cm or more),10cases of nodular goiter. Using immunohistochemicalmethod to detect confirmed thyroid pathological tissues IMP3and CD44v6expression level, test results with Biosens Digital Imaging System carries on thequantitative analysis v1.6professional image analysis software, choose integraloptical density (IOD) as evaluation parameters. At the same time, the data was analyzed by software SPSS17.0ResultsIMP3is mainly expressed in tumor cell cytoplasm and nucleus (or), was in brown.IMP3IOD overall expression in each case are shown in table1; IMP3IOD in eachgroup the difference was statistically significant (F=357.82, P <0.05); IMP3expression in the PTC is higher than in benign group, the difference was statisticallysignificant (P <0.05); IMP3in PTC comparison between those with and withoutlymph node metastasis group, the difference was statistically significant (P <0.05).CD44v6expression mainly in tumor cell membrane, was in brown. Generalexpression of CD44v6in groups of IOD is shown in table2; The differences in theIOD of CD44V6in each group was statistically significant (F=273.29, P <0.05);CD44v6expression in the PTC is higher than in benign group, the difference wasstatistically significant (P <0.05); CD44v6PTC comparison between those with andwithout lymph node metastasis group, the difference was statistically significant (P <0.05). IMP3and CD44v6expression in the PTC into positive correlation (r=0.905, P<0.05). This paper further analysis IMP3and CD44v6in thyroid papillary carcinomatwo types (that is, a papillary thyroid follicular carcinoma and follicular thyroidpapillary carcinoma), the differences between IMP3, CD44v6no statisticallysignificant difference between the two (T=0.78, P>0.78, T=0.96, P>0.96).Conclusions1IMP3and CD44v6, which were positively correlated in thyroid papillary carcinoma,are closely related to the occurrence and development of thyroid papillary carcinoma.2IMP3and CD44v6can be used as the diagnosis of thyroid papillary carcinoma, theevaluation indicators to distant metastasis and prognosis.
