Objective:1. To detect the expression of insulin-like growth factor II m RNA binding protein 3(IMP3), transforming growth factor(TGF)-β1 in cervical intraepithelial neoplasia, and analysis the relationship between them. Clarify the mechanisms responsible for cervical cancer progression and metastasis.2. To investigate the relativity between clinicopathologic significance and these proteins in cervical squamous cell carcinoma. Then, provide some evidences for early diagnosis, treatment and prognosis of cervical carcinoma.3. To discuss the expression which before and after the therapy of radiation or chemotherapy, and hope to establish the cure standards.4. Use the tumor-bearing mice to research the change of IMP3 and TGF-β1 before and after the therapy. which is helpful for targeted therapy.Methods:1.We selected 200 cervical tissues cases, 40 cases of cervical carcinoma which before and after the therapy, 40 cases of cervical carcinoma without therapy, 60 cases of HSIL,30 cases of LSIL,30 cases of cervicitis were used as controls. We used immunohistochemical EV method to detect the expression of IMP3,TGF-β1 in these cervical tissues.2. Detected the proliferation inhibition rate With different concentrations of cisplatin or different adiological dose by CCK-8, explored the best concentrations or adiological dose.3. Used Western-blot method to detect the protein’s performances of IMP3,TGF-β1 in tumor-bearing mouse’s tumor tissues.4. Used SPSS13.0 statistical software for statistical analysis on the experimental data, the inspection level of α=0.05.Results:1.(1) Positive immunohistochemical stain for IMP3 was identified in the most of carcinoma cases(86.3%), HSIL cases(78.3%), some LSIL cases(33.3%), it’s negative in both cervicitis(P<0.05).The expression was concerned with tissue differentiation, clinical stage, invasiveness and lymph node metastasis(P<0.05), had nothing to do with the age(P>0.05).(2) The positive expression rate of TGF-β1 in cervical carcinoma was higher than other tissues(P<0.05), it’s slao concerned with tissue differentiation, clinical stage, invasiveness and lymph node metastasis(P<0.05), but not with age(P>0.05).2. There was a positively relationship between the two proteins(r=0.265,P<0.05).3. The positive expression rate of the two proteins could be declined by radiation or chemotherapy, there was a significant difference(P<0.05),4. The results of CCK-8 told us: The medium effect dose of cisplatin was 5ug/ml continue 72 h, and radiation was 4Gy.5. Western blot analysis showed that both the therapy of radiation or chemotherapy could dedline the expression of IMP3 and TGF-β1 in tumor-bearing mice’s tumor tissues, there was a big relevancy between the two proteins.Conclusion:1. The expression of IMP3 was increased by the severity of cervical disease. Then, we conjectured that IMP3 could prompted the inevitability of intraepithelial neoplasia to carcinoma.Thus, We could use it as a tumor marker for earlier diagnose.2. The expression of IMP3 and TGF-β1 was closely related with tissue differentiation and clinical stage, we could say that the two of them take part in tissue differentiation and invasiveness.3. The closely related of the two proteins provided the basis for revealed the transmission of TGF-β1, and clear the nonsmads path in particular. IMP3 joined in as one of regulatory factors.4. Because of the lower expression after therapy, we could use the two proteins to judge the efficacy of some new cure methods, it was assisted to the individualized treatments.5. Simulated the animal Models of cervical cancer, then, maked a experimental basis for proven mechanisms of cervical cancer and find the best treatment options.
