ObjectiveTo explore the possible association between the single nucleotide polymorphisms (SNPs) of phosphodiestierase(PDE) and bipolar disorder,and look for biological markets.Methods1. According to the Tenth Revision of the International Statistical Classification of Diseases and Related Health Problems (ICD-10) that diagnostic criteria of bipolar disorder;432patients were included in the han patients (including male182cases, female250cases), in the same zone chosed569people of han healthy controls (male322cases, female247cases);The study respectively compared the gene type and allele distribution difference in the two groups, and the correlation analysis with bipolar disorder.2. Using fluorescence quantitative polymerase chain reaction(PCR) technology, detect the2SNPs of PDE4Ars7256672, rs1051738.3. Choosing17.0for windows software SPSS for data processing. Adopt goodness-of-fit chi-square test, analysis whether genotypes, allele frequency accord with Hardy-Weinderg balance laws; adopt χ2inspection to compare the cases and controls genotypes and alleles frequency; adopt The level of significance for p=0.05.Results1.Two SNPs site testing results Detecting DNA samples of Han people shows that408patients、569health controls for rs1051738,and432patients、481health controls for rs7256672.The two sites conform to Hardy-Weinberg(p>0.05).2. Comparing genotype and allele frequency in case group and control group2.1Comparing the PDE4A rs1051738genotype and allele frequency in case group and control group: the case group AA, AC and CC three genotype frequency for0.12,0.08and0.80.and the healthy control for0.02,0.19and0.79.It is found that CC type of the case group is obviously higher than control group.There are significant differences(χ2=58.962, df=1,p<0.001). The case group: A allele frequency for0.16, C allele frequency is0.84;The control group: A allele frequency for0.12, C allele frequency for0.88;two groups of allele frequency compared that case group C allele frequency is higher than the control group. there are significant diferences (χ2=8.319, df=1,p=0.004).2.2Comparing the PDE4A rs7256672genotype and allele frequency in case group and control group: the case group GG, GT and TT three genotype frequency for0.18,0.50and0.32.and the healthy control for0.20,0.48and0.32.There are no significant differences(χ2=0.956,df=1,p=0.620). The case group: G allele frequency for0.43, T allele frequency is0.57;The control group: G allele frequency for0.45, T allele frequency for0.55;two groups of allele frequency compared that there are no significant diferences (χ2=0.580,df=1,p=0.446).3. Comparing genotype and allele frequency in Males and females for the case grouprs1051738the males AA, AC and CC three genotype frequency for0.12,0.09and0.79.and the females for0.13,0.07and0.80.There are no significant differences(χ2=0.388, df=1,p=0.824).The males group: A allele frequency for0.17, C allele frequency is0.83;The females group: A allele frequency for0.16, C allele frequency for0.84; there are no significant differences (χ2=0.050, df=1,p=0.823).rs7256672the males GG, GT and TT three genotype frequency for0.18,0.53and0.29.and the females for0.18,0.48and0.34.There are no significant differences(χ2=1.970, df=1, p=0.373). The males group: G allele frequency for0.45, T allele frequency is0.55;The females group: A allele frequency for0.41, T allele frequency for0.59; there are no significant differences (χ2=0.983, df=1,p=0.321).4. Comparing genotype and allele frequency in the cese group and control group for malesrs1051738:There are significant differences in the genotype frequency (χ2=35.777, df=1,p<0.001).and no differences in the allele frequency(χ2=3.235, df=1,p=0.072).rs7256672:there are no significant differences in the genotype and allele frequency(p>0.05).5. Comparing genotype and allele frequency in the cese group and control group for femalesrs1051738:there are significant differences in the genotype and allele frequency(p<0.05).rs7256672:there are no significant differences in the genotype and allele frequency(p>0.05).Conclusions1. There is association between the PDE4A rs1051738and bipolar disorder,and it maybe is the functional site for PDE4A.2. There is no evidence to prove that there have association between the rs7256672and bipolar disorder.
