Object:Bipolar disorder(BP) is a common disease in psychiatry,wih a highrate of morbility, mortality, recurernce rate and disability rate.The most commonsymptoms of the disease is recurrent attacks of mania and depression,which bringsheavy burden to the family and society and becomes one of the most importantproblem to the mental health on the world.With a better understanding of bipolardisorder and molecular biology,more and more evidences have shown that bipolardisorder is a complex hereditary disease involved in a lot of factors.In recentyears,studies on bipolar disorder have been focused on susceptibility genes andgenetic polymorphisms. Now people begain to realize that cytokines play a importantrole in the pathogenesis of BP.Researchers found that inflammation may happened inmany common comorbiditis with BP such as type2diabetes,obesity and Metabolicsyndrome. IL-1,IL-4,IL-10,TNF-α,IFN and TGF are the common cytokinesinvolved in inflammatory reation,among which IL-1,TNF are known asproinflammation cytokines and IL-4,IL-10are anti-inflammation cytokines.There areevidences show that these cytokine have gene polymorphisms.In our research weinvestigated the association between IL-1B,IL-4gene polymorphisms and bipolardisorder susceptibility by TaqMan quantitative polymerase chain reaction(PCR) tofind the possible correlation between them.At the same time we want to explore theway of using gene diagnosis in bipolar disorder.Method:We recruit138patient with bipolar disorder and143health controls.All of the patient are conformed with BP and meet the diagnostic criterion ofICD-10and CCMD-3,with no physical sicknesses and inherited diseases.Healthcontrols must have no phychiatric disorders and immune-system diseases,with nofamily history to the above diease,and no other physical sicknesses.2ml peripheralvenous blood of all the subjects are draw-off in the morning before breakfast and thenanticoagulated by EDTA.DNA genes of the objects were isolated with kit.TaqMan Fluoroqenic quantitative PCR was used to detect the polymorphism of IL-1-511A/G(rs16944) and IL-4+33T/C(rs2070874).Result:The two sites are in line with the Hardy-Weinberg equilibrium in thecase and control group(P>0.05).The phenoype frequencies of IL-1B-511A/G isstatisticaliy difference between case and control group, whie allele frequencydifferences were not statistically significant. Genotype frequencies and allelefrequency differences were not statistically significant with IL-4+33T/C. Per locusexposed to the risk allele genotype distribution difference was no statisticalsignificance. Association between genotype frequencies and allele frequencies andBD susceptibility was not found by the logistic regression analysis adjusted for sexand age with wild-type homozygous allele as a reference.Conclusion:IL-1B-511A/G polymorphism might be associated with thesusceptibility to BD patients in the Chinese Han population.
