Objective:To study and compare the effects of sesame on NF-κB level in rabbit aortic endothelial cells, and to establish the antiatherogenic mechanism of sesamin.Methods:Separated and cultured rabbit aortic endothelial cells, after successful passage logarithmic growth phase cells used in the experiment, the experiment cells divided into four groups randomly:control group, ox-LDL group, PDTC+ox-LDL group, Sesamin+ox-LDL group. RT-PCR was used to detect rabbit aortic endothelial cells in mRNA levels of NF-κB.Results:MTT results showed that the concentration of40,60,80μg/mL sesamin comparison with the control group, there were significant differences P<0.05.60μg/mL sesamin which there was a significant,difference in comparison with other concentrations of sesamin. RT-PCR results showed that RNA expression of NF-κB in the ox-LDL group was observably higher than the control group, there was a evident difference (P<0.05), RNA expression of NF-κB in the sesamin group and the PDTC group was observably lower than the ox-LDL group, there were evident difference (P<0.05), which the sesamin group and the PDTC group had s evident difference (P<0.05)Conclusions:Sesamin and PDTC can inhibit mRNA expression of NF-κB, sesamin inhibition was greater than PDTC.Sesamin can delay the formation of atherosclerosis, as anti-atherosclerotic provide a new direction. |
